核因子-κB在慢性髓细胞白血病急变期前后的变化 被引量:1

摘要 目的探讨核因子-κB(NF—κB)在慢性髓细胞白血病(CML)急变期前后的表达及意义。方法选择2011年2月~2013年2月于新疆喀什人民医院血液科住院治疗的25例慢性髓细胞白血病慢性期(CML—CP)(CML—CP组)、16例慢性髓细胞白血病急变期(CML—BC)患者(CML—BC组)、30例缺铁性贫血或巨幼细胞性贫血患者(对照组)为研究对象。采用凝胶迁移或电泳迁移率实验检测三组人骨髓细胞中NF-κB的表达。结果对照组患者的NF—KB活性测定均为阴性.CML—CP组也仅4例患者检测到NF—KB活性增强,CML—BC组14例患者的NF—KB活性增强。CML—BC组患者NF—κB活化率明显高于CML—CP组及对照组,差异均有统计学意义(P〈0.05或P〈0.01);CML—CP组活化率高于对照组,差异有高度统计学意义(P〈0.01)。CML—BC组患者NF—KBmRNA表达水平(22.91±0.15)明显高于对照组(0)及CML—CP组(11.19±0.03),差异有统计学意义(P〈0.05)。结论与慢性髓细胞白血病慢性期比较.急变期NF-κB活性增强。表达增加,可作为慢性髓细胞白血病急性病变的重要标志物。 Objective To study the changes of the NF-kB in acute transformation phase of chronic myelogenous leukemia and its significance. Methods From February 2011 to February 2013, 25 cases of chronic myelogenous leukemia (CML-CP) (CML-CP group); 16 cases of chronic myelogenous leukemia patients in the acute transformation phase (CML-BC) (CML-BC group); and 30 patients with iron deficiency anemia or gigantic young cell anemia (control group) in Department of Hematology of First People's Hospital of Kashgar District were selected as the research objects. Expression of NF-KB in bone marrow cells of the three groups were tested by EMSA. Results Activity determination of NF-KB in the control group were all negative, NF-KB activity of 4 patients in the CML-CP group were increased, and NF-KB of 14 patients in the CML-BC group were activity. Activation rate of the CML-BC group was higher than that of CML-CP group and the control group, the differences were statistically significant (P 〈 0.05 or P 〈 0.01); activation rate of the CML-CP group was higher than that of the control group, the difference was statistically significant (P 〈 0.01). The expression level of the NF-KB mRNA in the CML-BC group (22.91±0.15) was significantly higher than that of control group (0) and CML-CP group (11.19±0.03), the differences were statistically significant (P 〈 0.05). Conclusion Compared with the chronic phase of chronic myelogenous leukemia, activity of NF-KB in the CML-BC phase was enhanced, the expression was increased, so NF-κB can be used as an important marker in predicting of CML-BP of chronic myelogenous leukemia.
