摘要
目的探索RNA干扰与^(131)I-免疫脂质体联用在胶质瘤疾病治疗中的潜在价值。方法在合成p GPU6-GFP-Neo-EGFRv III-shRNA表达载体的基础上,结合核素内放射性^(131)I免疫脂质体,进行细胞实验探索其对胶质瘤细胞U251增殖凋亡的影响。结果成功构建p GPU6-GFP-Neo-EGFRv III-shRNA载体;同时细胞实验结果显示,shRNA和^(131)I免疫脂质体联用有效抑制胶质瘤细胞U251增殖和促进其凋亡,抑制率达70%。结论 shRNA结合^(131)I免疫脂质体能高效地促凋亡和抑制细胞增殖,为其在胶质瘤治疗中的应用奠定了初步基础。
Objective To investigate the potential value of RNAi combined with ^131I-immune liposome in glioma treat- ment. Methods Structuring the vector harboring EGFRv III shRNA expression cassettes, used in cell experiments with ^131I-im- mune liposome, to study the effect on proliferation and apoptosis of U251 cells. Results shRNA combined with ^131I-immune lipo- some targeted to EGFRv III was able to effectively inhibit the proliferation and apoptosis of U251 cell. The inhibition rate was 70%. Conclusion shRNA combined with ^131I-immune liposome can efficiently promote apoptosis and inhibit cells proliferation, which laid a preliminary foundation for its application in the treatment of gliomas.
