结核分枝杆菌Rv0867c基因的生物信息学分析 被引量:3

摘要 目的应用生物信息学软件分析结核分枝杆菌Rv0867c及其编码的复苏促进因子A(resuscitation-promoting factor A,Rpf A)的结构功能及抗原表位。方法从NCBI数据库获取Rv0867c的基因信息;应用ProParam预测Rpf A蛋白的理化性质;采用SignalP 4.0及TMHMM分析其信号肽及跨膜区;利用在线分析Expasy工具分析蛋白二级结构,建立蛋白的三级结构模型;利用ABCpred、BepiPred 1.0Server以及SYFPEITHI、NetMHCIIpan 3.1Server分析蛋白的B细胞及T细胞抗原表位。结果 Rpf A蛋白具有407个氨基酸残基,该蛋白N端存在信号肽序列,具有跨膜结构。二级结构中无规卷曲约占63.14%,结构疏松。三级结构同源建模与Rpf B具有高度相似的序列。Rv0867c具有潜在的B细胞抗原表位46-57、61-74、108-123、307-320及85-98等,T细胞抗原表位82-90、337-345、167-175、207-215及263-271等。结论生物信息学预测Rpf A具有良好的抗原表位,是诊断及治疗结核的潜在候选因子。 Objective To use bioinformatics software to analyze the Rv0867 cgene from Mycobacterium tuberculosis and predict the structure and function of resuscitation-promoting factor A(Rpf A). Methods Basic information on the gene Rv0867 cwas obtained from the NCBI database.ProParam was used to predict the physical and chemical properties of Rpf A.SignalP 4and TMHMM were used to analyze the signal peptide and transmembrane region.The secondary and tertiary structure of Rpf A were modeled with Expasy.ABCpred,BepiPred,SYFPEITHI,and NetMHCIIpan 3.1Server were used to analyze epitopes. Results The Rv0867 cgene of Mycobacterium tuberculosis codes for the protein Rpf A,which consists of 407 amino acid residues.The protein has signal peptides and an apparent transmembrane structure.Random coils account for 63.14 % of the structure of the Rpf A protein,and it has a loose structure.Rpf A has a tertiary structure similar to that of Rpf B.There are potential B cell epitopes at or near amino acids 46-57,61-74,108-123,307-320,and 85-98.There are potential T cell epitopes at or near amino acids 82-90,337-345,167-175,207-215,and263-271. Conclusion Bioinformatics predicted that Rpf A might be a potential target protein for research on diagnosis of tuberculosis.
