摘要
目的观察丹槐银屑浓缩丸对豚鼠实验性银屑病病理改变及免疫系统调节的影响,探讨其作用机制。方法豚鼠40只,采用随机数字法随机分为模型组、维A酸组、丹槐低、中、高剂量组各8只,采用普萘洛尔溶液涂于豚鼠双耳背侧4周法造模,造模型成功后,维A酸组采用阳性药维A酸乳剂于皮损双耳涂药,丹槐低、中、高剂量组用丹槐银屑浓缩丸灌胃,模型组不处理。测定治疗前后豚鼠静脉血T淋巴细胞、红细胞、白细胞、单核细胞数量,测定血清总蛋白、血红蛋白、白蛋白含量,采用酶标双抗体夹心ELISA法测定肿瘤坏死因子(TNF-α)、干扰素(IFN-γ)、细胞因子白介素-1(IL-1)、IL-2、IL-6、IL-8等水平。结果模型组静脉血T淋巴细胞、单核细胞、白细胞数量均有不同程度升高,血清总蛋白、白蛋白及血红蛋白无明显变化。维A酸组静脉血T淋巴细、单核细胞,白细胞数量明显降低,对体液免疫系统有抑制作用,豚鼠血清总蛋白、白蛋白及血红蛋白含量降低,与模型组比较差异有统计学意义(均P<0.05)。丹槐中、高剂量组治疗后静脉血T淋巴细、单核细胞,白细胞数量明显降低,与本组治疗前及模型组比较差异有统计学意义(均P<0.05),其血清总蛋白、白蛋白及血红蛋白有降低均势,但与治疗前比较差别不大(均P>0.05),且丹槐中、高剂量组比较差别不大(均P>0.05)。治疗前后模型组白介素TNF-α、IFN-γ及IL-1、IL-2、IL-6、IL-8等均保持在较高水平,说明白介素炎性因子是引起动物皮肤发生银屑病样病理改变的基础。维A酸组白介素TNF-α、IFN-γ及IL-1、IL-2、IL-6、IL-8等均明显降低,与本组治疗前及模型组比较差异有统计学意义(均P<0.05)。丹槐中、高剂量组与维A酸组类似,其作用稍弱于维A酸,在治疗后TNF-α、IFN-γ及IL-1、IL-2、IL-6、IL-8等均明显降低,与本组治疗前及模型组比较差异有统计学意义(均P<0.05)。丹槐中、高剂量组比较差别不大(均P>0.05)。模型组、维A酸组、丹槐低、中、高剂量组涂普萘洛尔的表皮均有类似于人类银屑病样改变,造模后出现明显的皮损等病理学特征,治疗前5组动物病理检查均有表皮角质层过度角化,棘层变厚、钉突延长,真皮层出现中重度炎细胞浸润,角质层血管炎、内有小脓肿等银屑病性改变。治疗后维A酸组皮损消失,表皮角化现象明显减少,几无炎细胞浸润、表皮钉突及小脓肿消退。丹槐中、高剂量组皮损面积降低,但病理检查仍有部分动物表皮有角化现象,角质层血管炎、小脓肿等银屑病性较模型组及治疗前明显好转。结论丹槐银屑浓缩丸可在不影响体液免疫的情况下降低实验性银屑病豚鼠细胞免疫反应,抑制TNF-α诱导细胞因子与IFN-γ之间的级联反应,对白介素IL-1、IL-2、IL-6及IL-8等细胞因子介导的炎症反应亦有明显的抑制作用,可减轻炎症反应,具有与维A酸类似药理作用。
Objective:To observe the effect of dhyxnsw on regulation of guinea pig experimental psoriasis pathological changes and the immune system,to explore its mechanism of action.Methods:40 guinea pigs,were randomly divided into model group,A retinoic acid group,Dan Huai low,medium and high dose group.The propranolol solution was coated on the back side of the guinea pig ears for 4 weeks to model.And after successful modeling,A retinoic acid group with positive medicine a agent in lesions of binaural medicine,Dan Huai low,middle and high dose groups,with dhyxnsw gavage,compared model group without treatment.Before and after treatment,the venous blood T lymphocytes of guinea pig red blood cells,white blood cells,mononuclear cell number determination,determination of serum total protein,hemoglobin,albumin,tumor necrosis factor were determined by enzyme labeled by double antibody sandwich ELISA(TNF-α),interferon(IFN-γ),cytokine interleukin(IL-1),IL-2,-1 IL-6,IL-8 level.Results:dhyxnsw significantly decreased in the high dose group of Dan Huai,T lymphocytes,monocytes,TNF-α,IFN-γ,IL-1,IL-2,interleukin IL-6 and IL-8 secretion and synthesis also significantly inhibited,compared with the model group(P 0.05),the difference was statistically significant,but Dan Huai in high dose group,the above indexes between two groups had no significant difference(P 0.05).Conclusion:dhyxnsw decreased cellular immunity in guinea pig psoriasis does not affect humoral immune reaction in the experimental conditions,the inhibition of TNF-α induced cascade reaction between cytokines and IFN-γ,also had an obvious inhibitory effect on the inflammatory reaction of interleukin IL-1,IL-2,IL-6 and IL-8 mediated by cytokines,reducing inflammation the reaction,with pharmacological action and similar A retinoic acid.
