摘要
目的探讨原花青素(procyanidins,PC)对局灶性脑缺血大鼠细胞凋亡和鼠脑组织TNF-α、HSP70和含量的影响。方法选用健康SD大鼠30只,雌雄各半,随机分为假手术组、模型组、PC50 mg/kg组、100 mg/kg组和200 mg/kg组,共5组。24 h后进行神经功能评分,并断头取脑,制备组织匀浆,检测大鼠脑细胞凋亡率、肿瘤坏死因子-α(TNF-α)和热休克蛋白70(HSP70)含量。结果模型组神经功能评分和大鼠脑细胞凋亡率、TNF-α高于假手术组,差异有显著性(P<0.05);模型组大鼠脑组织HSP70含量低于假手术组,差异有显著性(P<0.05)。PC 50 mg/kg组、100 mg/kg组和200 mg/kg组的神经功能评分和大鼠脑细胞凋亡率、TNF-α低于模型组,差异有显著性(P<0.05);HSP70含量高于模型组,差异有显著性(P<0.05)。结论 PC对缺血性脑损伤具有保护作用,其保护作用机制可能与其抑制鼠脑细胞凋亡,并抑制TNF-α的生成,增加HSP70的生成有关。
Objective To observe the effect of procyanidins on neural cell apoptosis and the content of TNF-α,HSP70 in rat with focal cerebral ischemia. Methods 30 healthy SD rats, half male and half female, were randomly dividedinto the sham-operative group, focal cerebral ischemia model group and procyanidins group that was subdivided into 3 groupswith 50 m g /k g 100 mg/kg and 200 mg/kg procyanidins respectively. The neurological deficit score of rats was assessed andrat brain cell apoptosis rate,the content of TNF-α and HSP70 in brain tissue were measured 24 h after focal cerebral ischemia.Results The neurological deficit score of sham-operative group was better than that of model group, the percentage ofapopotisis cells and the content of TNF-α were lower in sham-operative group than in model group,the content of HSP70 was higher in sham-operative group than that of model group (P 〈0. 05) . The neurological deficit score were better in theseprocyanidins preconditioning group with 50 mg/kg, 100 mg/kg and 200 mg/kg than that of model group(P 〈0. 05). Thepercentage of apopotisis cells and the content of TNF-α were lower in these procyanidins preconditioning group with50 mg/kg, 100 mg/kg and 200 mg/kg than in model group,the content of HSP70 were higher in these procyanidins preconditioninggroup than that of model group (P 〈0. 05). Conclusions Procyanidins that has been given prior to the focal cerebralischemia plays a neuroprotect role by decreasing the content of TNF-α and apopotisis, increasing the content of HSP70.
