Correlation between fetuin-A gene polymorphism(rs4917,rs4918) and atherosclerosis cerebral infarction

目的探讨辽宁地区汉族人群胎球蛋白A（fetum-A）基因的rs4917 和rs4918 多态性与动脉粥样硬化性脑梗死（ atherosclerotic cerebral infarction,ACI）的相关性.方法纳人ACI患者1 6 7 例作为病例组,1 3 5 例同期体检的健康人作为正常对照组.通过改良多重连接酶反应技术（ improved multiplex ligation detection reaction, IMLDR）检测fetuin-A 基因rs4917 和rs4918 多态性.应用多因素Logistic回归分析研究基因多态性与A C I的相关性.结果病例组与对照组rs4917 和rs4918 基因型分布差异有统计学意义（ P 〈 0 . 0 5 ） ;病例组患者rs4917 的T 等位基因频率和rs4918 的G 等位基因频率与对照组相比差异有统计学意义（ P 〈 0 . 0 1 ） ; 进一步枸建显性模型和隐性模型,在隐性模型下两组差异有统计学意义（ P 〈 0. 01 ） ; 在平衡了 ACI各危险因素后,L0glstlc回归分析显示在隐性模型下 rs4917 及 rs4918 仍与 ACI 易感性相关（OR 3 .0 3 9 , P = 0 . 0 1 7 ,9 5 % CI 1 .2 1 7 ~ 7 . 5 8 9 ） .结论 fetuin-A 基因rs4917 及rs4918 多态性与辽宁地区汉族人群ACI易感性相关;rs4917 的T /T 基因型及M 9 1 8 的G/ G 基因型可能是辽宁地区汉族人群A CI的危险因素.

Objective To investigate the correlation between fetuin-A rs4917 and rs4918 gene polymorphism andatherosclerosis cerebral infarction （ACI） in Han population of Liaoning area. Methods A total of 167 patients with ACIwere enrolled as the case group and 135 healthy subjects over the same period were enrolled as the control group. The improvedmultiplex ligase chain reaction （IMLDR） was used to perform genotyping for fetuin-A gene rs4917 and rs4918. Andthe correlation between the gene polymorphism and ACI was analyzed with multivariate logistic regression analysis. ResultsGenotype distribution of rs4917 and rs4918 in the case group was different（P 〈0. 05） from the control group; T allelefrequency in rs4917 and G allele frequency in rs4918 in the case group were different （P 〈0. 01） from the control group;when build the dominant and recessive model, there was difference（ P 〈0. 01） between the two groups in recessive model;After adjusting for risk factors of ACI,multiple logistic regression analysis showed rs4917 and rs4918 still have correlationwith susceptibility of ACI （ OR = 3. 039, P = 0. 017,9 5 % C/： 1. 217 ~ 7. 589）. Conclusion Fetuin-A gene rs4917 andrs4918 polymorphisms have correlation with susceptibility of ACI in Han population of Liaoning area. The T/T genotype ofrs4917 and G/G genotype of rs4918 may be risk factors for ACI susceptibility.
