血清miR-103和Dicer1表达及在结直肠癌中的诊断价值

Detection of serum miR- 103 and Dicer1 and diagnostic significance for colorectal cancer patients

目的:探讨结直肠癌患者血清miR-103和Dicer1表达情况,分析miR-103联合Dicer1检测在结直肠癌诊断中的应用前景。方法:72例经病理确诊的结直肠癌患者为实验组,72例健康体检者为对照组。应用实时定量PCR法检测实验组和对照组血清miR-103和Dicer1表达水平。分析两种生物标志物联合诊断结直肠癌的价值。结果:与对照组相比,miR-103在实验组患者血清中表达水平显著上调(P<0.01),Dicer1水平显著下调(P<0.05)。miR-103表达水平与结直肠癌临床分期、肿瘤转移情况以及Dicer1水平密切相关(P均<0.05)。miR-103与Dicer1联合在结直肠癌检测的敏感度和特异度最高可达82.1%和95.6%,Logistic分析受试者工作特征曲线(ROC)曲线下面积(AUC)为0.754。结论:miR-103高表达与结直肠癌分期、转移及Dicer1表达关系密切,与Dicer1联合有望成为结直肠癌的早期诊断指标。

Objective： To investigate the expression of miR- 103 and Dicer1 in serum of colorectal cancer patients,and to analyze the application of miR- 103 combined with Dicer1 detection in the diagnosis of colorectal cancer. Methods： All 72 cases of colorectal cancer patients diagnosed by pathology as the experimental group,72 cases of healthy persons as the control group. The expression levels of miR- 103 and Dicer1 were detected by real- time quantitative PCR assay. Results： Compared with the control group,the expression of miR- 103 in the experimental group was significantly higher（ P〈0. 01）,and the serum Dicer1 level was significantly lower in the experimental group（ P〈0. 05）. The expression level of miR- 103 was closely related to the clinical stage of colorectal cancer,tumor metastasis and the level of Dicer1（ all P〈0. 05）. The sensitivity and specificity of miR- 103 and Dicer1 combination in colorectal cancer were 82. 1% and 95. 6%,respectively. The area under the receiver operating characteristic curve（ ROC） was 0. 754（ AUC）. Conclusion： The high expression of miR- 103 was closely related to the clinical stage,metastasis and Dicer1 expression of colorectal cancer,and the combination of Dicer1 and was expected to be an early diagnostic target for colorectal cancer.
