IL-10基因载体对大鼠血管平滑肌细胞增殖和球囊损伤后新生内膜形成的影响

摘要 目的探讨白细胞介素-10(IL-10)基因载体对自发性高血压大鼠血管平滑肌细胞(SHRVSMC)增殖和球囊损伤后新生内膜形成的影响。方法成功构建慢病毒IL-10基因载体。自发性高血压大鼠(SHR)随机分为4组:正常对照组、模型组、损伤+空载体组(空载体组)、损伤+慢病毒IL-10基因治疗组(IL-10基因治疗组)。大鼠颈动脉球囊损伤模型建立后,取出颈总动脉下段1/3行HE染色,计算机图像分析仪测量血管壁(中膜)/腔面积比(wall area/lumen area,W/L)。颈总动脉段上1/3检测金属基质蛋白酶-12(matrix metalloproteinase,MMP-12)的信使核糖核酸(messenger ribonucleic acid,mRNA)表达。结果慢病毒IL-10基因治疗组W/L比值比模型组和空载体组明显减小;IL-10基因治疗组MMP-12的mRNA相对表达量比模型组和空载体组明显减小。结论 IL-10基因治疗能抑制大鼠体内血管平滑肌细胞增殖和球囊损伤后新生内膜形成,是有效的再狭窄治疗手段。 Objective To investigate the effect of interleukin-10(IL-10)gene vector on the proliferation of vascular smooth muscle cells(SHRVSMC)and the formation of intimal hyperplasia after balloon injury in spontaneously hypertensive rats.Methods Lentivirus IL-10 gene vector were successfully constructed.Spontaneously hypertensive rats(SHR)were randomly divided into four groups:normal control group,model group,damage+empty vector group(empty vector group),damage+lentivirus IL-10 gene therapy group(IL-10 gene therapy group).After the establishment of rat carotid artery balloon injury model,remove the carotid artery under paragraph 1/3was removed for HE staining,the walls of blood vessels in the film)/lumen area ratio(wall area/lumen area,w/L)were measured by using computer image analyzer,and MMP-12 expression of matrix metalloproteinase mRNA was detected in the carotid artery segment of the common carotid artery in 1/3.Results The ratio of W/L to lentivirus IL-10 in the treatment group was significantly decreased than that in the model group and empty vector group,and the relative expression of MMP-12 mRNA in the IL-10 gene therapy group was significantly smaller than that in the model group and empty vector group.Conclusion IL-10 gene therapy can inhibit the proliferation of vascular smooth muscle cells and the formation of intimal hyperplasia after balloon injury in rats,which is an effective method for the treatment of restenosis.
