摘要
以椰子油为原料,对催化癸酸与椰子油酯交换制备富含中碳链甘三酯的脂肪酶进行筛选,对脂肪酶Lipozyme RM IM、脂肪酶Lipozyme TL IM、脂肪酶Novozyme 435和猪胰脂酶催化癸酸与椰子油酯交换反应效果进行比较,脂肪酶Lipozyme RM IM比其他几种脂肪酶的催化效果好。在底物物质的量之比(油/酸)为1∶3、加酶量(以总底物质量计)为4%、加水量(以酶质量计)为10%条件下,考察不同温度、不同时间脂肪酶Lipozyme RM IM催化癸酸与椰子油酯交换制备富含中碳链甘三酯的影响。结果表明:该条件下催化癸酸反应后,产物中中碳链甘三酯含量为37.46%,癸酸含量为35.72%,相比于原料椰子油分别增长了22.53个百分点和30.98个百分点。
Enzymatic transesterification is an important method for the preparation of functional lipids from fatty acids and triglycerides,which is widely used in the study of structural lipid preparation. The present study was to screen the lipase in the catalyzed synthesis of triglycerides enriched in medium carbon-chain(MCT),which was prepared by transesterification of coconut oil and capric acid. The effect of Lipozyme RM IM,Lipozyme TL IM,Novozyme 435 and porcine pancreas lipase on the capric acid and MCT contents in the catalyzed products were investigated. The results showed that the lipase RM IM had better catalyzed effect than other three lipases. The effect of catalysis temperature and time on the MCT contents in the lipase RM IM catalyzed transesterification of coconut oil and capric acid was also examined under the following conditions: the substrate mole ratio of oil to fatty acid of 1 ∶3,enzyme load of 4%(W/W of total substrate) and 10% water(W/W of lipase). It was concluded that under the optimal conditions,the content of MCT and capric acid were 37.46% and 35.72%,which increased by 22.53 percent points and 30.98 percent points respectively compared to the raw material.
