摘要
建立了养殖水中孔雀石绿和结晶紫及其代谢物残留的分析方法。方法以二氯甲烷为提取溶剂,提取液经酸性氧化铝萃取柱净化,外标法定量。方法使用液相色谱仪紫外检测器同时双波长检测与荧光检测器串联,优化了检测波长,孔雀石绿和结晶紫及其代谢物独立检测,提高定性、定量的准确性。添加0.005~0.500mg/kg浓度水平时,孔雀石绿和结晶紫及其代谢物加标回收率在79.5%-89.2%之间,相对标准偏差(RSD)为5.2%-7.1%之间。方法检出限(S/N=3)为0.5~2.0gg/kg,定量限(s,N=10)为2.0-5.0μg/kg。该方法简单快速,准确可靠,可用于基层检测站的日常抽样快速检测工作。
The paper presents a method of analysis of aquaculture water of malachite green and crystal violet and its residues. Methods using dichloromethane as extraction solvent, extraction liquid by acidic alumina extraction column purification, using external standard method. Method using liquid chromatography with ultraviolet detector and dual wavelength detection with fluorescence detector connected in series and to optimize the detection wavelength, malachite green and crystal violet and their metabolites independent detection and improve the accuracy of qualitative and quantitative. Add 0.005-0.500 mg/kg concentration and recovery rate between the 79.5 %-89.2 % malachite green and crystal violet and their metabolites and, relative standard deviation(RSD)was between 5.2 %-7.1%. The detection limit(S/N=3)for 0.5-2.0 g/kg(S/N=10), the quantitation limit is 2.0-5.0 g/kg. The method is simple, rapid, accurate and reliable, can be used for daily sampling for rapid detection of primary inspection station.
