克唑替尼的合成进展

摘要 克唑替尼是MET/ALK激酶双靶标抑制剂,可能成为ALK融合蛋白呈阳性的非小细胞肺癌患者的潜在靶向药物。从2007年立项到2011年FDA批准上市,仅用了4年时间。现综述了克唑替尼的合成进展,为该药的进一步研究提供参考。 Crizotinib, a dual MET/ALK inhibitor, was approved by the U.S. Food and Drug Administration(FDA)for the treatment of anaplastic lymphoma kinase(ALK)-rearranged non-small cell lung cancer(NSCLC)in August 2011, merely 4 years after the first publication of ALK-rearranged NSCLC. The paper reviews the synthetic methods of crizotinib.
