参附注射液对老年大鼠心肌缺血再灌注损伤的保护作用 被引量:5

摘要 目的探讨参附注射液对老年大鼠心肌缺血再灌注损伤的保护作用及其机制。方法 45只18月龄Wistar雄性大鼠随机分为假手术组、模型组和预处理组,每组15只。预处理组腹腔注射参附注射液10mg/kg,1次/d,术前连续给药7d,末次给药2h内进行模型制作,术后停药。假手术组及模型组给予同容量生理盐水腹腔注射。采用结扎左冠状动脉前降支制备缺血再灌注模型。电镜观察心肌超微结构改变、免疫组织化学检测心肌组织NF-κB、超氧化物歧化酶(superoxide dismutase,SOD)和丙二醛水平。结果模型组的相对光密度值(0.335±0.124 vs0.270±0.112、0.293±0.102,P<0.05)、丙二醛水平[(72.68±5.23)mmol/g vs(41.89±2.01)mmol/g、(48.52±7.14)mmol/g,P<0.01]明显高于假手术组、预处理组,SOD水平[(62.32±3.10)U/g vs(116.54±10.21)U/g、(95.64±9.12)U/g,P<0.01]明显低于假手术组和预处理组。结论参附注射液可以抑制NF-κB活性,减少氧自由基损伤,从而减轻心肌组织缺血再灌注损伤。 Objective To study the role of of Shen Fu Injection in protection of aged rats against myocardial I/R injury and its mechanism.Methods Forty-five Wistar male rats aged 18 months were randomly divided into sham operation group,model group,pretreatment group(15in each group).The animals in pretreatment group were injected with Shen Fu Injection(10mg/kg,once a day,7days in a roll).A rat myocardial I/R injury model was established by injecting Shen Fu Injection into the abdominal cavity.The myocardial ultrastructure of rats was observed by elctron microscpy.The serum NF-κB,SOD and MDA levels were measured with immunohistochemical staining.Results The relative optical densisty and serum MDA level were significantly higher whereas the serum SOD level was significantly lower in model group than in sham operation group and pretreatment group(0.335±0.124 vs 0.270±0.112,0.335±0.124 vs0.293±0.102,P〈0.05;72.68±5.23mmol/g vs 41.89±2.01mmol/g,72.68±5.23mmol/g vs 48.52±7.14mmol/g,P〈0.01;62.32±3.10U/g vs 116.54±10.21U/g,62.32±3.10U/g vs 95.64±9.12U/g,P〈0.01).Conclusion Shen Fu Injection can inhibit the NF-κB activity and reduce the free radical damage,thus alleviating the myocardial I/R injury.
