当归四逆汤对心肌缺血大鼠心电图及心肌酶谱的影响 被引量:3

Effect of Danggui Sini Dectoction(当归四逆汤, DGSND)on Electrocardiogram and Myocardial Enzyme Spectrum of Myocardial Ischemia in Rats

摘要 目的:观察当归四逆汤对垂体后叶素(Pit)所致心肌缺血大鼠心电图及心肌酶谱的影响。方法:采用雄性Wistar大鼠腹腔注射垂体后叶素Pit(30 U/kg)造成急性心肌缺血损伤模型,观察各组大鼠注射Pit前10 min,及注射5、15、30 min后的心率、Ⅱ导联心电图变化,并检测血清乳酸脱氢酶(LDH)、磷酸肌酸激酶(CPK)活性。结果:注射Pit 5 min后,DSD高、低剂量组及阳性对照组心率均高于模型组,但差异均无统计学意义(P>0.05);注射Pit15 min、30 min后,DSD高、低剂量组及阳性对照组心率均高于模型组(P<0.01或P<0.05)。注射Pit后,各组大鼠心电图均出现不同程度的波形改变,主要表现为ST段向上偏移,T波高耸,心律失常等。DSD高、低剂量组及阳性对照组ST段、T波变异性程度均低于模型组。模型组LDH、CPK活性均高于空白对照组(P<0.01),DSD高、低剂量组及阳性对照组LDH、CPK活性均明显低于模型组,差异均有统计学意义(P<0.05或P<0.01)。结论:当归四逆对心肌缺血模型大鼠心电图ST段、T波、心率有改善作用;能降低LDH、CPK的活性,具有保护心肌细胞的作用。 Objective: To observe the effect of Danggui Sini Dectoction on Electrocardiogram and myocardial enzymes in rats myocardial ischemia caused by Pituitrin (Pit). Methods: With male Wistar rats, acute myocardial ischemia model caused by intraperitoneal injection of Pit (30 PU/kg); The heart rate, I1 electrocardiogram (ECG) and sermn lactate dehydrogenase (LDH), creatine kinase (CPK) were observed 10 rain before injection of pit of rats, and 5, 15 rain and 30 rain after the injection. Results: 5 rain after injection of pit, the DGSND high and low dose groups and positive control group showed higher heart rate than model group, but the diflbrence was not statistically significant (P〉0.05); and 15 rain and 30 min after the injection of pit, the DGSND high and low dose groups and positive control group showed higher heart rate than model group (P〈0.05 or P〈0.05). After in- jection of Pit, ECG of rats in each group showed different degree of waveform changes, mainly for the ST seg- ment upward shift, T wave high, arrhythmia and so on. DGSND high and low dose groups and positive control group showed less change of ST segment and T wave variability than model group. Model group showed higher LDH and Cpk activity than the blank control group (P〈0.01); the DGSND high and low dose groups and positive control group showed lower LDH and Cpk activity than the model group, and the difference was statistically sig- nificant (P〈0.05 or P〈0.01). Conclusion: Danggui Sini Dectoction can improve the electrocardiogram ST segment, T wave and heart rate of myocardial ischemia model in rats, decrease the activity of LDH and CPK, and protect the cardiac muscle cells.
