摘要
人类对肿瘤发病机制的认识经历了一个漫长的进程,从过去单一的物理致癌、化学致癌、病毒致癌、突变致癌系统上升到多步骤、多因素综合致癌理论,其中癌基因、抑癌基因、DNA损伤修复基因的活性在此过程中起到了关键的作用。RAD18是一种DNA损伤激活的E3泛素连接酶,在肿瘤细胞的DNA损伤修复中起重要作用。RAD18在多种肿瘤细胞中表达水平升高明显,在放化疗产生抵抗的肿瘤细胞中表达进一步升高,提示RAD18与肿瘤的发生相关,并可作为克服耐药性的一个潜在靶点。
It has been a long process to understand the pathogenesis of tumor, from physical factors, chemical factors, viruses and mutation to the multi-step and multi-factor process theory, and the activity of oncogenes,tumor suppressor genes and DNA damage repair genes play key roles in the process. RAD18, a ubiqultin ligase ( E3 ), has been known to play an important role in the repair of DNA damage in tumor cells. Several studies found that the expression of RADlg significantly increased in various tumor cells,and further increased when developing resistance to chemotherapy or radiotherapy, suggesting that RADI8 is related to the occurrence of tumor and provide a new therapeutic target to overcome drug resistance.
