摘要
目的检测寻常性银屑病患者外周血髓源性抑制细胞(MDSCs)和Th17以及IL-17A的表达水平,并分析MDSCs与Th17细胞的相关性。方法选取120例寻常性银屑病患者(进展期58例,稳定期62例)为研究组,以40例健康者为正常对照组,运用流式细胞术(FCM)检测各组研究对象的外周血MDSCs和Th17细胞频率;采用酶联免疫吸附法(ELISA)检测血清中IL-17A的表达水平,并分析MDSCs与Th17细胞的相关性。结果活动组中MDSCs的百分比明显低于稳定组,而Th17细胞频率及IL-17A含量均明显高于稳定组,差异有统计学意义(P<0.01)。稳定组与健康对照组MDSCs细胞百分比比较,差异无统计学意义(P>0.05),而活动组和稳定组的Th17细胞及IL-17A的含量均高于正常对照组(P<0.05)。MDSCs百分比与Th17细胞百分比及IL-17A的含量均呈现负相关性,差异有统计学意义(P<0.05)。结论临床活动性寻常性银屑病患者外周血中的MDSCs呈现低水平,且与Th17细胞百分比及IL-17A的含量呈现负相关性。提示外周血中的MDSCs和Th17细胞可能呈相互拮抗作用,并且共同参与寻常性银屑病的发生、发展。
Objective To detect the myeloid-derived suppressor cells (MDSCs) in the peripheral blood of patients with psoriasis vulgaris and to analyze its correlation with Th17 ceils and expression of interleukin-17A (IL-17A). Methods One hundred and twenty cases of psoriasis vulgaris, including 58 cases of progressive psoriasis and 62 cases of stable psoriasis, and 40 healthy controls were enrolled in this study. The peripheral blood MDSCs and Th17 cells were detected using FCM. ELISA was used to detect the expression levels of IL-17A. The correlation between MDSCs and Th17 cells was analyzed. Results The percentages of MDSCs in patients with progressive psoriasis were significantly lower than that in stable psoriasis ( P 〈 0. 01 ). The percentages of Th17 cells and the expression levels of IL-17A in the progressive group were significantly higher than that in the stable group (P 〈 0. 01 ). The percentages of MDSCs were no statistical differences between the stable and the normal controls (P 〉 0. 05 ), but the percentages of Th17 cells and the expression levels of IL-17A in the progressive group and the stable group were higher than those in the normal controls (P 〈 0. 05). The percentage of MDSCs was negatively correlated with the percentages of Th17 cells and levels of IL-17A (P 〈 0. 05 ). Conclusion The percentages of MDSCs in peripheral blood are low in the patients with progressive psoriasis vulgaris, and are negatively correlated with the percentages of Th17 cells and IL- 17A levels, suggesting that MDSCs and Th17 cells in peripheral blood antagonize each other, involving the pathogenesis of psoriasis vulgaris.
