中国对虾(Fenneropenaeus chinensis)肌肉蛋白提取物对致敏小鼠体内酶的影响

Alterations of Enzymes Activities in ICR Mice Sensitized by Muscle Protein Extracts from Chinese Shrimp(Fenneropenaeus chinensis)

摘要 本文研究腹腔注射中国对虾(Fenneropenaeus chinensis)肌肉蛋白提取物前后小鼠体内几种酶因过敏反应而产生的变化。以小鼠的肝脏、肾脏、脾脏、血清为试验样本,对其体内的超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)、谷胱甘肽-S-转移酶(GST)、一氧化氮合酶(NOS、iNOS、c NOS)、磷脂酶A2(PLA2)以及钙离子ATP酶(Ca2+-ATPase)的活性、浓度或m RNA相对表达量进行分析。测定结果显示:腹腔注射不同剂量虾蛋白粗提物后,各组小鼠血清中s Ig E水平显著增加,说明过敏模型已成功建立。过敏个体内肝、肾、脾和血清中SOD、POD、CAT、GSH-Px、GST、NOS(i NOS)活性明显提高,PLA2浓度也显著增加,而肝、肾、脾内Ca2+-ATPase m RNA相对表达量下降。本研究结果为中国对虾蛋白动物过敏的评价及反应机理提供一定理论依据。 The alterations of several enzymes activities in ICR mice were studied by intraperitoneal injection of muscle protein extracts from Chinese shrimp(Fenneropenaeus chinensis). The activities of SOD, POD, CAT, GSH-Px, GST,NOS(i NOS) and the concentration of PLA2 as well as the relative m RNA expression level of Ca2+-ATPase in the liver,kidney, spleen and sera were analyzed. The results showed that the s Ig E levels in the serum of mice were significantly increased after the shrimp protein extracts injection, indicating that the allergic model of mice was successfully established. Besides, the activities of SOD, POD, CAT, GSH-Px, GST and NOS(i NOS) in the liver, kidney, spleen and sera were remarkably enhanced in sensitized mice, so as the concentration changes of PLA2. However, the m RNA expression of Ca2+-ATPase was decreased after sensitization. Our results may provide some evidence for evaluation of allergic reactions in mouse model and better understanding the mechanism of allergy of Chinese shrimp extracts.
