摘要
在骨化二醇产生菌自养假诺卡菌(Pseudonocardia autotrophica)突变株SIIA243-924的发酵液中发现一个骨化二醇的结构类似物,分别采用液-液萃取、硅胶柱色谱和高效液相制备色谱等方法提取分离,得到纯度为97.5%的目标化合物。并进一步通过UV、MS、1H NMR、13C NMR及1H-1HCOSY等光谱分析,结合骨化二醇相关谱图,确定该化合物的结构为(5Z,7E)-9,10-开环胆甾-5,7,10(19)-三烯-(26-甲基)-3β,25-二醇,即26-甲基-25-羟基维生素D3。
A cacifediol analogue was found from the broth of a mutant strain Actinomycetes SIIA243-924, which was a producing strain for cacifediol. The target compound was isolated by means of liquid-liquid extraction, silica gel column chromatography and preparative HPLC, with a purity of 97.5 %. Its chemical structure was confirmed by UV, MS, tH NMR, 13C NMR and 1H-1HCOSY, which was (5Z,7E)-9,10-secocholesta-5,7,10 (19)-triene-(26-methyl)-313,25- diolmonohydrate, named as 26-methyl-25-hydroxy vitamin D3.
