摘要
为了提高彩色小麦麸皮花色苷的纯化效率,保证花色苷组分的稳定,以AB-8大孔树脂为填料,采用层析法对彩麦麸皮花色苷初提样品进行纯化,对影响大孔树脂纯化花色苷效果的几个主要条件进行了优化。结果表明,花色苷样品溶液最佳上样浓度为75 mg/m L,最佳上样流速为2 m L/min,最大上样体积为320 m L,以1 m L/min的流速用60%的乙醇作为洗脱溶剂洗脱效果较好,250 m L的洗脱溶剂可将花色苷基本洗脱完全。经纯化后的花色苷样品纯度为4.23 mg/g,与纯化前相比,纯度提高了9.04倍。
The color wheat bran anthocyanin was purified by AB-8 macroporous resin to improve its purification efficiency and guarantee the stability of anthocyanins component.The purification condition of the macroporous resin was optimized.The results of static adsorption and desorption experiments showed that the best sample initial concentration of anthocyanin crude product was 75 mg/m L and the best sample flow rate was 2 m L/min;The largest sample volume was 320 m L and the effect of 1 m L/min flow rate with 60% ethanol as elution solvent elution was good.Anthocyanins could be completely eluted by 250 m L basic elution solvent.The purity of anthocyanin samples after purification was 4.23 mg/g.When compared with the beginning,the purity increased 9.04 times and purification effect was better.
