吡格列酮对戊四氮致痫大鼠Toll样受体、核因子-κB表达的影响 被引量:1

摘要 目的研究过氧化物酶增殖物激活受体(PPAR)γ激动剂吡格列酮对癫痫大鼠海马组织Toll样受体(TLR)4及核因子(NF)-κB表达的影响。方法建立雄性SD大鼠腹腔注射PTZ诱导癫痫模型,根据组别给予不同剂量PTZ连续灌胃,RT-PCR法检测各组大鼠海马区TLR4及NF-κB含量,免疫荧光检测TLR4及NF-κB蛋白表达。结果通过RT-PCR及免疫组化检测,癫痫模型组(PTZ)大鼠脑海马TLR4、NF-κB含量比正常对照组(NC组)增高(P<0.05);PTZ干预低、中、高剂量组(PL、PM、PH组)的TLR4、NF-κB表达量较PTZ组减低(P<0.05),且随着PTZ干预剂量增加而调低,并于PH组显著下降(P<0.05)。结论戊四氮致痫大鼠海马区TLR4及其下游信号NF-κB表达增加,PPARγ激动剂吡格列酮能够通过降低TLR4及NF-κB表达,从而减轻癫痫发作对神经系统的炎性损伤,这可能是其对神经细胞保护作用的潜在机制之一。
