摘要
目的研究双黄连粉针剂与盐酸左氧氟沙星注射液对小鼠细胞色素P4502E1(CYP2E1)酶活性的影响。方法将28只小鼠按照体重随机分为4组:中药组(腹腔注射双黄连粉针剂3.7 g·kg^(-1)·d^(-1))、西药组(腹腔注射左氧氟沙星注射液1.0 g·kg^(-1)·d^(-1))、联合组(腹腔注射双黄连粉针剂3.7 g·kg^(-1)·d-1+左氧氟沙星注射液1.0 g·kg^(-1)·d-1)、空白组(腹腔注射0.9%Na Cl)。连续给药7 d后,眼内眦取血,用双抗体酶联免疫法测定小鼠血浆中CYP2E1酶的活性。结果与空白组的酶活性(27.79±1.16)pg·m L^(-1)比较,西药组与联合组的酶活性分别为(22.84±0.75),(21.34±0.59)pg·m L^(-1),组间比较差异有统计学意义(P<0.01)。中药组的酶活性为(24.85±1.10)pg·m L^(-1),与空白组比较,差异无统计学意义(P>0.05)。结论联合组对CYP2E1活性有显著抑制作用,该抑制作用是由左氧氟沙星引起的。
Objective To evaluate the effect of Shuanghuanglian Powder for injection and levofloxacin hydrochloride injection on the activity of cytochrome P450 2El (CYP2E1). Methods Twenty eight male mice were randomly divided into 4 groups, the Chinese herbs group (Shuanghuanglian Powder, 3.7 g ·kg^-1·d^-1 ), the antibiotics group ( levofloxacin hydrochloride injection,1.0 g ·kg^-1·d^-1), the combination group (Shuanghuanglian Powder 3.7g·kg^-1·d^-1 and levofloxacin hydrochloride injection 1.0 g·kg^-1·d^-1) and the blank group. After intraperitoneal administration of drugs for 7 days, the activity of serum CYP2E1 was detected to enzyme linked immunosorbent assay. Results Compared to the activity of CYP2E1 with (27. 79 ± 1.16 ) pg·mL^-1in blank group, the activity of CYP2E1 in the antibiotics group with (22. 84 ±0. 75) pg·mL^-1 and combination group with(21.34 ±0. 59) pg ·mL^-1 notably decreased (P 〈0. 01 ). However, there was not notably different between Chinese herbs group (24.85 ± 1.10)pg ·mL^-1and blank group (P 〉 0. 05 ), and there also had no notably difference between antibiotics group and combination group (P 〉 0.05 ). Conclusion CYP2EI activity can be inhibited notably by levofloxacin but not Shuangtmanglian. Levofloxaein plays an important role in the inhibition of CYP2E1 in combination group.
