联合应用衣霉素增加顺铂诱导人大细胞肺癌H460细胞凋亡

The combined application of tunicamycin and cisplatin-induced apoptosis in human large-cell lung cancer H460 cells

摘要 目的探讨顺铂联合衣霉素对人大细胞肺癌H460细胞生长增殖的作用。方法 H460细胞在体外培养,MTT方法检测在不同浓度顺铂单独组及联合作用24h对细胞活性的影响。用Western blot法检测H460细胞内cleaved caspase-3和cytochome C的表达水平。结果 MTT结果显示顺铂能抑制H460细胞的增殖并呈现剂量依赖性,同时顺铂可以引起H460细胞中凋亡相关蛋白cleaved caspase-3和cytochome C的表达水平明显的升高,差异有统计学意义(P<0.05)。联合衣霉素可以进一步增加顺铂引起的H460细胞的增殖抑制作用,同时进一步增加凋亡相关蛋白cleaved caspase-3和cytochome C的表达,差异有统计学意义(P<0.05)。结论衣霉素可以增加H460细胞对顺铂的敏感性,从而提高顺铂治疗大细胞肺癌的疗效。 Objective To explore the effect of cisplatin in combine with tunicamycm on the growth of human large cell lung cancer H460 ceils. Methods H460 cells were cultured in vitro and MTT assay was used to examine the cell viability of cells treated by cisplatin alone or in combine with tunieamycin for 24 h. Western blot was applied to examine the expression levels of cleaved caspase-3 and eytochome C in H460 cells. Results MTT results revealed that cisp]atin could inhibit the growth of H460 cells in concentration dependent manners and increase the expression of apoptotic related proteins cleaved caspase-3 and cytochome C, and the difference was statistically significant (P〈0.05). Combined with tunicamycin can further inhibit the growth of H460 cells in- duced by cisplatin and up-regulate the expression of cleaved caspase-3 and eytochome C,and the difference was statistically signifi- cant(P〈0.05). Conclusion Tunicamycin can increase the cisplatin sensitivity of H460,which raise the anticancer effect of cisplatin to human large cell lung cancer.
