摘要
为了检测分析ADPN及其受体在鸡骨骼和成骨细胞上的表达情况,本研究提取蛋鸡龙骨和鸡成骨细胞总RNA,采用PCR定性分析鸡骨骼和成骨细胞中是否含有ADPN、脂联素受体1(Adiponectin receptors1,AdipoR1)和脂联素受体2(Adiponectin receptors2,Adi-poR2),实时荧光定量PCR分析蛋鸡龙骨和成骨细胞中AdipoR1和AdipoR2 mRNA的相对表达量。结果:鸡龙骨和成骨细胞中表达AdipoR1和AdipoR2;AdipoR2 mRNA含量显著高于AdipoR1 mRNA。研究表明:AdipoR1和AdipoR2在鸡骨骼和鸡成骨细胞中表达,且提示在骨骼中ADPN可能主要与AdipoR2结合,通过内分泌的形式影响鸡骨骼代谢。
To detect whether or not the ADPN and its receptor expressed in chicken bone and osteoblast cells, the total RNA and protein were extracted from the keel bone and osteoblasts of hens. The AdpioR1, AdpioR2, osteo- calcin and β-actin mRNA of chicken keel bone and osteoblasts were detected by PCR. The level of osteocalcin, Ad- pioR1 and AdpioR2 mRNA of keel bone of hens and chicken osteocalcin were analyzed by Real-time PCR. The results showed that AdipoR1 and AdipoR2 expressed in the keel bone and osteoblasts of chicken. However, expression of ADPN couldnot be found in chicken keel bone and osteoblasts. And AdipoR2 had higher expressed value than Ad- pioR1. The results indicated that adiponectin might have effects on the chicken bone metabolism via the AdiopoR2 through endocrine pathways, and the regulation mechanism of adiponectin on chicken bone may be different with mammal.
