金黄色葡萄球菌肠毒素A的原核表达及鉴定

摘要 [目的]高效制备金黄色葡萄球菌肠毒素A(SEA)用于SEA快速检测相关产品的研究。[方法]通过原核表达的方法表达SEA,对其纯化方法进行研究,通过Western-blot和制备的SEA顺磁微粒免疫层析快速检测试纸条检测其免疫原性。[结果]SEA成功克隆入原核表达载体p ET22b(+),并在E.coil BL21(DE3)中得到有效表达,表达量达63.8%;采用低温诱导可获得可溶性的SEA,其表达量占总SEA的56.3%;可溶性SEA经Ni柱亲和层析一步纯化可得到纯度在95%左右的SEA,Western-blot检测显示所纯化的重组SEA具有SEA的免疫原性。[结论]研究构建的SEA表达载体能够高效、经济、简单的制备SEA,纯化的SEA可用于食物中毒中SEA快速检测相关产品的开发,具有一定的应用价值。
