载淫羊藿素多孔PLGA/TCP联合微骨折术修复兔膝关节软骨缺损的初步观察 被引量:3

摘要 目的研究载淫羊藿素多孔聚乳酸-羟基乙酸共聚物(PLGA)/磷酸三钙(TCP)材料联合微骨折术修复兔膝关节软骨缺损中骨与软骨结合的特性。方法 24只兔子制造膝关节软骨缺损动物模型,随机分成空白组、对照组、实验组,每组8只,在软骨缺损处进行微骨折处理,分别不植入材料、植入PLGA/TCP材料和植入携载淫羊藿素的PLGA/TCP材料,6个月后进行CT评估及大体观察。结果 CT显示实验组膝关节材料构建骨形态结构良好,对照组骨形态结构稍差,空白组良好骨支架结构丧失;大体观察显示实验组修复生软骨组织色泽接近正常,对照组软骨色泽稍灰暗,空白组示无软骨修复,大体评分结果显示实验组优于对照组及空白组(P<0.05),对照组优于空白组(P<0.05)。结论携载淫羊藿素的PLGA/TCP材料修复兔膝关节软骨缺损其支架结构与骨整合良好,软骨大体观察效果良好,为进一步探索其修复效果提供初步依据。 Objective To explore the characteristics of the PLGA/TCP scaffolds incorporating icariin in repairing the defect of knee cartilage of the rabbits.Methods The model of cartilage defect was created on the patellar groove.They were drilled by micro-fracture and divided into blank group,control group and experimental group,which received no scaffold,PLGA/TCP scaffolds and the the PLGA/TCP scaffolds incorporating icariin.They were sacrificed after 6 months. CT was used to explored the bone scaffold/three-dimensional structure and the gross observation was used to evaluated the articular cartilage.Results The CT scan showed they had whole and complete structure of knee in the experimental group and worse structure in the control group,the blank group had lose the whole structure.The gross observation showed the experimental group kept fresh color,and grey in the control group,the defect was kept in the blank group. The experimental group was better than that in control group and blank group (P〈0.05),and control group was better than blank group (P〈0.05).Conclusion The PLGA/TCP scaffolds incorporating icariin can keep good structure in repair- ing the defect of knee cartilage.The gross observation of the articular cartilage in the experimental group is best.It pro- vides primary basis for exporing the repair effect father.
