小细胞肺癌患者化疗前后血清ProGRP、CEP350、NSE含量评估及其与疾病预后的关系 被引量:9

Assessment of serum ProGRP,CEP350 and NSE levels in patients with small cell lung cancer before and after chemotherapy and their relationship with disease prognosis

摘要 目的:研究小细胞肺癌患者化疗前后血清中胃泌素释放肽前体(ProGRP)、人中心体蛋白350(CEP350)、神经元特异性烯醇化酶(NSE)含量及其与疾病预后的关系。方法:选择97例小细胞肺癌患者作为研究的病例组,按照EP方案进行化疗;选择100例健康体检者作为研究的对照组。采集两组受试者的血清标本并测定ProGRP、CEP350、NSE含量,采集病例组患者的肿瘤组织、瘤旁组织并测定膜联蛋白A1(ANXA1)、Nm23-H1、YKL-40、AMPK相关蛋白激酶(ARK5)的mRNA含量。结果:病例组血清ProGRP、CEP350、NSE含量显著高于对照组(P<0.05);病理组中肿瘤直径超过4 cm、发生淋巴结和远处转移患者的血清ProGRP、CEP350、NSE含量较高(P<0.05);肺癌组织中ANXA1、YKL-40、ARK5的mRNA含量显著高于瘤旁组织(P<0.05)且与ProGRP、CEP350、NSE含量呈正相关,Nm23-H1的mRNA含量显著低于瘤旁组织(P<0.05)且与ProGRP、CEP350、NSE含量呈负相关;化疗后完全缓解(CR)、部分缓解(PR)、稳定(SD)患者的血清ProGRP、CEP350、NSE含量均低于化疗前(P<0.05),而进展(PD)患者化疗后的血清ProGRP、CEP350、NSE含量均高于化疗前(P<0.05)。结论:血清ProGRP、CEP350、NSE含量对小细胞肺癌的病情发展、化疗效果以及预后情况均具有评估价值。 Objective: To study the serum ProGRP,CEP350 and NSE levels in patients with small cell lung cancer before and after chemotherapy and their relationship with disease prognosis. Method: A total of 97 cases of patients with small cell lung cancer were selected as the pathology group of the study and received EP scheme chemotherapy; 100 cases of healthy subjects were selected as the control group of the research. Serum samples were collected from two groups to determine ProGRP,CEP350 and NSE levels,and the tumor tissue and peri-tumor tissue were collected from pathology group to determine the mRNA levels of ANXA1,Nm23-H1,YKL-40 and ARK5. Results: Serum ProGRP,CEP350 and NSE levels of pathology group were significantly higher than those of control group; serum ProGRP,CEP350 and NSE levels in pathology group of patients with tumor diameter more than 4 cm,lymph node metastasis and distant metastasis were higher; mRNA levels of ANXA1,YKL-40 and ARK5 in lung cancer tissue were significantly higher than those in peri-tumor tissue and were positively correlated with ProGRP,CEP350 and NSE levels,and mRNA level of Nm23-H1 was significantly lower than that in peri-tumor tissue and was negatively correlated with ProGRP,CEP350 and NSE levels; serum ProGRP,CEP350 and NSE levels in PR,CR and SD patients after chemotherapy were lower than those before chemotherapy,and serum ProGRP,CEP350 and NSE levels in PD patients after chemotherapy were higher than those before chemotherapy. Conclusions:Serum ProGRP,CEP350 and NSE levels can assess the progression of disease,effect of chemotherapy and prognosis of small cell lung cancer.
