摘要
目的:研究雷公藤多苷所致雄性幼鼠生殖损伤的可逆性。方法:第一部分:3~4周龄SD雄性幼鼠14只,随机分为空白组和多苷组,每组7只。空白组给予0.5%羧甲基纤维素钠(CMC-Na)灌胃,多苷组给予雷公藤多苷混悬液(9 mg·kg-1·d-1)灌胃,持续给药12周后停药4周。观察停药4周后大鼠睾丸组织的病理变化及睾丸组织雄激素受体(AR)蛋白的表达情况。第二部分:3~4周龄SD雄性幼鼠20只,随机分为空白组和多苷组,每组10只。各组大鼠灌胃药物与剂量及给药周期同"第一部分"。给药12周、停药4周后,将大鼠分笼喂养,每笼1只,按照1∶1比例放入成年雌性大鼠1只,合笼时间为2周,分3次合笼,观察雌鼠受孕率、仔鼠存活率及仔鼠发育情况。结果:1停药4周后,多苷组大鼠的睾丸曲细精管恢复良好,间距正常,细胞层次排列分明,无变性、水肿,间质细胞、精原细胞、精母细胞明显恢复,精子细胞及精子清晰可见。2停药4周后,多苷组大鼠睾丸组织AR蛋白的表达与空白组比较,差异无统计学意义(P〉0.05)。3合笼后,两组的雌鼠受孕数、受孕率、产仔数、离乳存活率比较,差异均无统计学意义(P〉0.05)。结论:雷公藤多苷对雄性幼鼠的生殖损伤作用具有可逆性恢复,且不影响大鼠的生殖能力。
Objective: To study the reversibility of reproductive damage induced by Tripterygium Wilfordii Hook. F( GTW) in the male adolescence rats. Methods: The part one: Fourteen SD male adolescence rats who were 3 ~ 4 weeks old were randomly divided into the blank control group and the GTW group,with 7 rats in each group. The rats in the blank control group were treated with 0. 5% carboxymethylcellulose sodium( CMC-Na) and the rats in the GTW group were treated with GTW( 9 mg·kg-1·d-1). The rats were continuously treated for 12 week,and then the drugs were stopped. Four weeks after drugs withdrawal,the testis pathological changes and the androgen receptor( AR) protein expression were observed. The part two: Twenty SD male adolescence rats who were 3 ~ 4 weeks old were randomly divided into the blank control group and the GTW group,with 10 rats in each group. The administration drugs and dosage and the treatment course were the same with"the part one". Four weeks after drugs withdrawal,the rats were allotted to cages,and each cage was put into an adult female rat at 1 ∶ 1 ratio to mate three times for 2 weeks. The pregnancy rate,the survival rate of offspring and the offspring development situation were observed. Results: 1 Four weeks after drugs withdrawal,the testis seminiferous tubules of rats in the GTW group were recovered,the interstitial cells were arranged in neat rows,the space was normal with no edema and denaturation.The mesenchymal cell,spermatogonia and spermatocytes were recovered and the spermoblast and sperm were seen clearly.2 Four weeks after drugs withdrawal,there was no statistical difference on the AR protein expression in the testis tissue between the blank control group and the GTW group( P 〉 0. 05). 3 There were no statistical differences on the number of pregnancy,the pregnancy rate,the number of offspring and the survival rate of offspring between the blank control group and the GTW group( P 〉 0. 05). Conclusion: The reproductive damage induced by GTW in the male adolescence rats has reversibility and has no influence on the reproductive ability of rats.
