加减薯蓣丸对血管性痴呆大鼠海马BDNF mRNA表达的影响 被引量:13

Effects of Modified Yam Pill on the Expression of Brain-derived Neurotropic Factor in the Hippocampus of Vascular Dementia Rats

摘要 目的:通过血管性痴呆大鼠模型,研究加减薯蓣丸对模型大鼠行为学及海马区脑源性神经营养因子(brain-derived neurotrophic factor,BDNF)表达的影响,探讨加减薯蓣丸干预血管性痴呆的可能机制。方法:采用改良双侧颈总动脉阻断法,将大鼠双侧颈总动脉分次结扎并剪断,选取造模成功大鼠分为模型组和中药组,并设假手术组,每组20只。中药组予加减薯蓣丸10 g·kg-1·d-1,其余2组用等量生理盐水。连续灌胃8周后,各组大鼠进行Morris水迷宫实验(定位航行实验、空间探索实验);取海马组织,应用荧光定量PCR检测海马区BDNF mRNA表达水平。结果:与假手术组比较,模型组大鼠的逃避潜伏期明显延长、穿越平台次数减少、游动速度下降(P<0.05),海马组织BDNF mRNA表达水平明显降低(P<0.05);与模型组比较,中药组大鼠逃避潜伏期缩短,穿越平台次数和平台象限停留时间延长(P<0.05),中药组大鼠海马区BDNF mRNA表达明显升高(P<0.05)。结论:通过改良的双侧颈总动脉阻断法造模,可有效模拟血管性痴呆的行为学下降;加减薯蓣丸方可提高海马区下降的BDNF mRNA表达水平,这可能与影响神经突触可塑性作用有关。 Objective: To explore the effects of modified yam pill on the behavior of vascular dementia rats and the expression of brain-derived neurotropic factor( BDNF) in the hippocampus,and discuss its possible mechanism on the intervention of vascular dementia. Methods: The rats model was established by modified method of cutting bilateral common carotid arteries. Established model rats were divided into the model group,traditional Chinese medicine group and sham operation group,20 rats in each group. The traditional Chinese medicine group was given modified yam pill at the dose of 10 g·kg-1· d-1,and other groups were given equivalent normal saline. After treatment for 8 weeks,Morris water maze experiments( positioning navigation experiment and space exploration experiment) were carried out in each group. The hippocampus were taken and the mRNA expression of BDNF was estimated by quantitative fluorescent PCR. Results:Compared with the sham group,the escape latency of rats in model group prolonged and crossing platform number reduced,swimming speed decreased( P 〈 0. 05) and the mRNA expression of BDNF in hippocampus significantly decreased( P 〈 0. 05); compared with the model group,the escape latency of rats in traditional chinese medicine group shortened,crossing platform number and platform residence time prolonged( P 〈 0. 05),and the mRNA expression of BDNF in hippocampus significantly increased( P 〈 0. 05). Conclusion: Modified yam pill may increase the expression of BDNF in the hippocampus and thus play its role against vascular dementia,which may be related to the nerve synapses.
