巴西橡胶树JAZ家族基因的克隆和表达分析 被引量:5

Cloning and Expression Analysis of JAZ Gene Family of Hevea brasiliensis

摘要 茉莉酸是调控橡胶树创伤反应及产排胶的重要激素,JAZ蛋白是茉莉酸信号途径中重要的负调控因子。目前,巴西橡胶树中已报道7个JAZ基因。本研究结合Genebank中已有的橡胶c DNA序列和本实验室转录组测序数据,通过电子克隆的方法获得5个橡胶JAZ基因全长序列,分别命名为HbJAZ3、HbJAZ4、HbJAZ5、HbJAZ6和HbJAZ12,并通过RT-PCR技术成功扩增得到HbJAZ3、HbJA5、HbJAZ6、HbJAZ12等4个基因的编码区。最后采用定量RT-PCR技术分析发现,4个橡胶JAZ基因在橡胶不同组织中的表达有明显,以及其在伤诱导和茉莉酸处理后的差异表达,发现4个橡胶JAZ基因的表达都受到茉莉酸及创伤处理的影响,说明这些JAZ蛋白参与橡胶茉莉酸调控的创伤反应,本研究为阐明橡胶产排胶调控机理打下基础。 Jasmonic acid (JA) is one of phytohormone involved in wounding response and latex biosynthesis and drainage. JAZ proteins are the most important repressors of plant JA signaling pathways. Totally, 7 JAZ proteins have been reported in Hevea brasiliensis so far. In this work, combined with the cDNA sequences deposited in GenBank and RNA-seq data of our lab, five full-length cDNA sequences of new JAZ gene were predicted by in silico procedure for H. brasiliensis, of which four genes were successfully amplified by RT-PCR and termed as HbJAZ3, HbJAZS, HbJAZ6, and HbJAZ12, respectively. Data of quantitative PCR technique showed that these genes were differentially expressed in different tissues and the wounding treatment and jasmonic acid treatment up regulated their expression, suggesting that JAZ gene involved in JA related wounding response. This work lays a foundation for clarification of molecular mechanism of latex regulation in H. b rasilie nsis.
