摘要
目的利用响应面设计法优化壳聚糖絮凝纯化丹参饮水提液工艺。方法在单因素研究的基础上,以液料比、壳聚糖加入量、絮凝温度和絮凝时间为考察因素,以透光率、固含物保留率和丹酚酸B保留率综合评分为评价指标,使用Box-Behnken设计实验方案,借助Design Expert 8.0.6b试验设计软件绘制响应面曲线图,优选最佳絮凝工艺。结果确定最佳絮凝工艺为液料比10.7 mL/g,壳聚糖加入量1.2 mL/g,絮凝温度51℃条件下处理1 h。结论响应面法优化壳聚糖絮凝纯化工艺稳定可行,除杂率高,有效成分损失量少。
Objective To optimize the flocculation technology of water-extraction solution from Danshen Yin with chitosan by response surface methodology. Methods The response surface model was designed by Design Expert 8.0. 6b program, and the best formulation was optimized on the basis of single factor analysis, with solid-liquid ratio, chitosan amount, flocculation temperature and flocculation time as the detecting factors, and the light transmission, solid content and salvianolic acid B retention rates as the evaluating indicators. Results The best flocculation conditions were as follows:solid-liquid ratio was 10. 7 mL/g with chitosan amount of 1.2 mL/g, and the temperature and time of flocculation was 51 ℃ and 1 h. Conclusion The flocculation clarification technology optimized by response surface method is feasible and stable with high rate of impurity removal and high retention rate of active ingredient.
