摘要
目的:探讨miR200c和E-Cad、ZEB1的靶向关系。方法:采用qRT-PCR检测子痫前期患者和正常妊娠者胎盘组织miR200c和E-Cad、ZEB1表达;western blotting检测E-Cad和ZEB1蛋白水平表达。结果:qRT-PCR结果显示,与正常妊娠者比较,子痫前期患者胎盘组织中miR200c的表达水平显著增高,E-Cad mRNA表达水平显著升高,ZEB1 mRNA表达水平显著降低,差异具有统计学意义(P<0.05)。western blotting结果显示,与正常妊娠者比较,子痫前期患者E-Cad蛋白水平显著升高,ZEB1蛋白水平显著降低,差异具有统计学意义(P<0.05)。结论:miR200c调控E-Cad和ZEB1的表达。
Objective: To investigate the relationship between miR200c, E -Cad and Zinc finger E - box - binding protein 1 (ZEB1) expression. Methods: The expression of ZEB1 and E - Cad and rniR200c in the placenta of patients with preeclampsia and normal pregnant women were detected by qRT - PCR, and the expression of E - Cad and ZEB1 protein was also detected by western blotting. Results: qRT - PCR results showed that the expression level of miR200c in placenta tissue of patients with preeclarnpsia was significantly higher than control group, and the expression of E - Cad rnRNA was significantly increased, and the expres- sion of mRNA ZEB1 was significantly decreased ( P 〈 0. 05 ). Western blotting results showed that the ex- pression level of E - Cad was significantly increased in patients with preeclampsia, and the expression level of ZEB1 was significantly decreased (P 〈0. 05). Conclusion: miR200c regulates the expression of ZEB1 and E - Cad.
