摘要
目的比较结蛋白(Desmin)在前列腺增生及前列腺癌中的表达差异,分析其与前列腺癌的关系。方法采用免疫化学SP法检测Desmin蛋白在前列腺增生组(30例)、前列腺癌组(30例)中的表达,并通过蛋白印迹Western-Blot法及实时定量Q-PCR法进行进一步验证,结合临床病理因素进行进一步分析。结果免疫组化SP法检测表明Desmin在BPH组和PCa组中的阳性表达率分别为73.33%(22/30)、43.33%(13/30),差异有统计学意义(P<0.05),且Desmin在前列腺癌中的阳性率与Gleason分级、病理分期呈负相关(P<0.05)。Western Blot法检测显示Desmin的蛋白表达水平在前列腺增生组明显高于前列腺癌组,差异均有统计学意义(P<0.05);Q-PCR法检测结果显示:在BPH组中Desmin mRNA表达水平高于PCa组,差异有统计学意义(P<0.05)。结论 Desmin在前列腺癌发生、发展中具有重要意义。
Objective To compare the differential expression of Desmin in benign prostatic hyperplasiaand prostate cancer,and to analyze the relationship between benign prostatic hyperplasia and prostate cancer. Methods We used immunohisto- chemical SP method to detect the expression of Desmin protein in benign prostatic hyperplasia(30 cases) and prostate cancer group (30 cases). And we further validated by Western-Blot method and real-time quantitative Q-PCR method, combined with clinical pathological factors for further analysis. Results Detectionof immunohistochemical SP method showed that the rate of positive ex-pression of Desmin in BPH and PCa group wererespectively73. 33% (22/30) ,43. 33% ( 13/30) , the difference was statistically significant( P 〈0. 05) ,and the positive rate of Desmin in prostate cancer was negatively correlated with Gleason grade and patho-logical staging( P 〈 0. 05 ). Western Blot method showed that the protein expression level of Desmin in hyperplasia of prostate gland group wassignificantly higher than that in the prostate cancer group,and the difference was statistically significant(P 〈0. 05) . The results of Q-PCR method showed that the expression level of mRNA Desmin in the BPH group was higher than that in the PCa group,and the difference was statistically significant(P 〈0. 05) . Conclusion Desmin is of great significance in the occurrence and development of prostate cancer.
