双石汤对高血压大鼠心肌肥厚的作用

Effects of Shuang Shi Tang on myocardial hypertrophy of reno-vascular hypertension rats

摘要 目的探讨双石汤对高血压大鼠心肌肥厚的作用及其机制。方法采用左肾动脉结扎法建立肾性高血压大鼠模型。实验分组:模型组、西药组、中药组、假手术组。中药组与西药组分别予双石汤及贝那普利灌胃,假手术组及模型组予等体积生理盐水。分别于术前、术后2周及用药4周、8周检测血压,用药8周后进行心脏超声检查,测定左室舒张末期内径(LVDd)及收缩末期内径(LVDs),左室舒张末期左室后壁厚度(LVPWd),左室收缩末期左室后壁厚度(LVPWs),射血分数(EF)。称取体重、全心重及左室重量,计算心重指数(CI)和左心室质量指数(LVMI),ELISA法检测血清中AngⅡ含量,PCR检测AT1R m RNA表达水平。结果与假手术组比较,中药组血压及LVDd、LVDs、LVPWd、IVSd降低(P<0.05),而EF升高(P<0.05);CI及LVMI降低(P<0.05);血清AngⅡ含量及AT1R的m RNA表达均下降(P<0.01)。结论双石汤能有效降低肾性高血压大鼠模型的血压,并通过抑制AngⅡ及其受体这一机制保护心肌。 Objective To investigate the effect of Shuang Shi Tang on myocardial hypertrophy of reno-vascular hypertension rats(RHR). Methods The models of renal hypertension were established by ligation of the left renal artery. The rats were divided into the model group, the herb group, the benazepril group and the sham operation group.During the 8 weeks of treatment, the herb group and the benazepril group were given intragastric administration every day, and the sham operation group and the model group were given equal volume of normal saline. The blood pressure was checked at 4 different periods. Transthoracic echocardiography was used to evaluate cardiac function. The left ventricle wet weight was recorded and normalized by body weight. The concentration of Ang II was examined by ELISA.The expression of AT1 R m RNA was examined by PCR. Results The herb depress the blood pressure obviously and reduced the CI and LVMI(P 0.05). LVDd, LVDs, LVPWd, IVSd of the herb group were lower, but EF were higher(P0.05) than those of the control group. The content of Ang II and the expression of AT1 R m RNA in the herb group were inhibited(P0.01). Conclusion Shuang Shi Tang has a function of controlling the blood pressure and inhibiting myocardial hypertrophy of RHR through the depression Ang II and AT1 R.
