氧气对嗜酸氧化亚铁硫杆菌磁性颗粒形成的影响

摘要 生活在高酸高浓度铁环境的嗜酸氧化亚铁硫杆菌其生物矿化作用可以形成胞内致密电子颗粒-磁性颗粒,其是一种具有极广应用潜能的生物纳米材料.为了探究氧气对嗜酸氧化亚铁硫杆菌生物矿化的影响,运用Real-time PCR技术,研究嗜酸氧化亚铁硫杆菌中2个与磁性颗粒形成相关的二价铁转运基因feoB、mpsA和三价铁转运基因tonB、ABC在不同转速下的差异表达,以及利用透射电子显微镜检测不同转速下磁性颗粒形成情况,结果发现氧气对基因的表达以及磁性颗粒的形成有一定影响,这为深入研究嗜酸氧化亚铁硫杆菌中磁性颗粒形成机理打下了基础. Acidithiobacillus ferrooxidans lives in aerobic environments where concentration of iron is extreme- ly high and the pH is low. It can synthesize intra-cellular electron-dense magnetite, called magnetic-particle, which can be extensively used as bio-nano-materials. In order to explore the influence of oxygen on magneto- some formation in Acidithiobacillus ferrooxidans, the different shaking frequency responses of feoB, mpsA, tonB and ABC gene were analyzed by using real-time PCR. Transmission electron microscopy was also used to analyze the magnetic-particle formed by this bacterium in different shaking frequencies. The result shows that the oxygen has some effect on the expression of the genes and the formation of the magnetic-particle, which lays the foundation for the deep research of the mechanism of the formation of magnetic-particle.
