槲皮素对丙泊酚麻醉术后小鼠认知功能的影响及其机制 被引量:2

摘要 目的研究槲皮素对丙泊酚麻醉诱导的术后认知功能障碍(POCD)的影响,并初步探讨其作用机制。方法槲皮素灌胃预处理10月龄小鼠2周后,进行丙泊酚麻醉和腹腔探查术,术后继续槲皮素灌胃处理1周(槲皮素组),同时设置对照组、麻醉组(丙泊酚麻醉)、手术组(丙泊酚麻醉后行腹腔探查术)。应用Morris水迷宫测定小鼠处理前后的定位航行能力和空间探索能力;分别应用实时PCR和Western blot检测小鼠海马晚期糖基化终末产物受体(RAGE)m RNA和蛋白表达水平。结果与对照组相比,麻醉组和手术组小鼠逃避潜伏期明显延长(P<0.05),原平台停留时间百分比明显下降(P<0.05),且手术组变化更为明显;槲皮素组与对照组相比,小鼠Morris水迷宫测试结果无明显差异。与对照组相比,麻醉组和手术组小鼠RAGE m RNA和蛋白表达水平均显著增高(P<0.05),而槲皮素组变化不明显。RAGE m RNA及蛋白表达水平与逃避潜伏期呈正相关,与原平台停留时间百分比呈负相关。结论槲皮素可改善丙泊酚麻醉诱导的POCD,该作用可能与抑制RAGE表达有关。 Objective To investigate the effect of quercetin on the postoperative cognitive dysfunction (POCD) induced by propofol anesthesia, and further explore the mechanism of this effect. Methods The mice were randomly divided into control group (normal feeding), anesthesia group (propefol), operation group (abdominal exploration after propofol anesthesia), and quercetin group. In quereetin group, the mice were pre- treated with quercetin by irrigation gastric for 2 weeks, then narcotized with propofol and underwent exploratory laparotomy, following with querce- tin treatment for 1 week. Morris water maze was used to determine the ability of positioning navigation and space exploration of mice. Advanced gly- cation end products receptor (RAGE) mRNA and protein expression levels were detected by real-time PCR and Western blot, respectively. Re- suits Compared with the control pup ,mice in anesthesia group and operation group exhibited significantly prolonged escape latency (P 〈 0.05 ), and the original platform residence time percentage decreased significantly (P 〈 0.05 ). In addition, the changes in the surgical group is more obvi- ous. Quercetin group mice showed no significant difference compared to control group in the Morris water maze test. RAGE mRNA and protein ex- pression levels of anesthesia group and operation group mice were significantly increased (P 〈 0.05) compared to control group, but not obviously in quercetin group. RAGE mRNA and protein expression levels were positively correlated with escape latency, but negatively correlated with the original platform residence time percentage. Conclusion Quercetin can improve the POCD induced by propofol anesthesia, which may be related to the inhibition of RAGE expression.
