摘要
目的:观察丹参地上部分SB-4-5075-0部位对给予不同预收缩刺激的大鼠肾主动脉的舒张作用。方法:采用DMT Myograph肌动描计系统记录离体血管张力。观察SB-4-5075-0部位(10-5~1 g·L-1)对基础状态和用K+(60 mmol·L-1)、PE(10-5 mol·L-1)、5-HT(10-5 mol·L-1)分别预收缩的内皮完整血管的舒张作用;观察SB-4-5075-0部位(10-5~1 g·L-1)对用K+(60 mmol·L-1)、PE(10-5 mol·L-1)、5-HT(10-5 mol·L-1)分别预收缩的去内皮血管环的舒张作用。高效液相检测SB-5075-0中主要有效成分。结果:在内皮完整的血管上,SB-4-5075-0部位对基础状态的血管没有舒张作用;在不同物质刺激预收缩的内皮完整及去内皮的血管上,SB-4-5075-0部位在(10-2~1 g·L-1)剂量区间范围内对用K+和5-HT预收缩的血管有显著舒张作用(P〈0.05,P〈0.01),对用PE预收缩的血管没有明显的舒张作用;SB-4-5075-0部位的主要有效成分为丹参素、原儿茶醛和咖啡酸。结论:SB-4-5075-0部位对正常血管无舒张作用,对K+和5-HT引起的血管收缩均有良好的舒张作用。
OBJECTIVE To investigate vasodilatory effects of SB-4-5075-0fractions of Salvia miltiorrhiza aerial parts on ex-vivo rat renal artery.METHODS By using DMT Myograph in vitro vascular tension meter system,muscular responses of rat renal arterial ring by SB-4-5075-0fractions were investigated.Diastolic function of intact or denuded endothelial rat renal artery were observed on basic state and stimulating state by K+(60 mmol·L-1),PE(10-5 mol·L-1)and 5-HT(10-5 mol·L-1).HPLC was applied to determine contents of SB-4-5075-0fractions of Salvia miltiorrhiza aerial parts,by using Diamonsil C18 column.RESULTS Within 10-5-1 g·L-1 dose range,SB-4-5075-0fractions showed no relaxing effects on basic state artery.Within 10-2-1 g·L-1 dose range,artery was relaxed after stimulating by K+or 5-HT(P〈0.05,P〈0.01).Within 10-2-1g·L-1 dose range,artery was not relaxed after stimulating by PE.Main components in SB-4-5075-0fractions were tanshinol,protocatechuic aldehyde and coffeic acid.CONCLUSION SB-4-5075-0fractions can relax artery stimulated by K+or 5-HT.
