普伐他汀对激素性坏死股骨头内Cbfa1表达的相关影响 被引量:4

摘要 目的分析普伐他汀对激素性坏死股骨头内Cbfa1表达的影响。方法将90只健康的成年新西兰白兔作为研究对象,随机分为3个组别,对照组、试验组、模型组各30只。试验组、模型组造成股骨头坏死模型,并对试验组灌服普伐他汀,对照组和模型组灌服蒸馏水。观察白兔的股骨头组织血管数量和骨陷窝缺损情况,采用荧光定量PCR法检测Cbfa1的表达水平。结果试验组白兔的血管数量为(8.4±1.6)条,血管直径为(9.8±1.2)μm,骨陷窝缺损率为(20.3±4.7)%,第8周、12周、16周股骨头内Cbfa1的表达水平分别为(0.531±0.074)、(0.717±0.079)、(0.885±0.083),均高于其他两组,且随着时间延长表达水平逐渐增高。结论普伐他汀能够促进坏死股骨头内Cbfa1表达,从而改善组织病理学,提示在激素性股骨头坏死的治疗中具有重要价值。 Objective To investigate the influence of pravastatin on Cbfa1 expression in steroid induced necrosis of femoral. Methods 90 healthy adult New Zealand white rabbits were randomly divided into 3 groups: the control group,the experimental group and the model group,with 30 rats in each group. In experimental group,model group,models of avascular necrosis of the femoral head were induced. And the experimental group was gavaged with pravastatin,the control group and model group were administrated with distilled water. We observed rabbit's bone tissue and blood vessel number and defect of bone and used fluorescence quantitative PCR method for detection of Cbfa1 expression levels. Results In the experimental group,the number of blood vessels were( 8. 4 ± 1. 6),vascular diameter was( 9. 8 ± 1. 2) μm,defect of bone lacuna rate was( 20. 3 ± 4. 7) %.At 8 weeks,1 2 weeks,1 6 weeks the in Cbfa 1 expression level of femoral head were( 0. 5 3 1 ± 0. 0 7 4) 、( 0. 7 1 7 ± 0. 0 7 9) 、( 0. 885 ± 0. 083). It is higher than the other two groups. Conclusion Pravastatin can promote the expression of Cbfa1 in avascular necrosis of the femoral head,so as to improve the pathological tissues. It is suggested that pravastatin has an important value in treatment of steroid induced femoral head necrosis.
