同时接种伪狂犬病活疫苗Bartha-K61株对嵌合载体猪瘟疫苗rAdV-SFV-E2免疫效力的影响 被引量:5

摘要 本实验室前期研究中构建了一种新型的猪瘟标记疫苗—腺病毒/甲病毒复制子嵌合载体猪瘟疫苗(rAdV-SFV-E2),并证实该疫苗具有与C株猪瘟疫苗相同的免疫效力。为评价该疫苗与伪狂犬病疫苗(Bartha-K61)联合免疫是否存在相互干扰现象,本研究将这两种疫苗联合免疫健康仔猪(n=5),同时设立单独免疫对照和阴性对照,免疫后利用CSFV强毒石门株进行攻击,并通过抗体反应、临床症状、病毒血症、病理学及组织病理学变化等指标对疫苗免疫效果进行评价。结果显示,rAdV-SFV-E2和Bartha-K61联合免疫后与单独免疫rAdV-SFV-E2均能够对致死性CSFV强毒的攻击提供完全保护(5/5存活),表现为无猪瘟特异性临床症状、无病毒血症、无病理变化和组织病理学变化。该研究证实,rAdV-SFV-E2和伪狂犬病疫苗Bartha-K61同时免疫对rAdV-SFV-E2的免疫效果无干扰现象。 In a previous study, we showed that an adenovirus-delivered, Semliki forest virus replicon-vectored vaccine (rAdV-SFV-E2) against CSFV was able to protect pigs against lethal classical swine fever virus (CSFV) challenge. In order to evaluate the efficacy of rAdV-SFV-E2 co-administered with pseudorabies vaccine Bartha-K61, four groups of 5-week-old pigs (n=5) were immunized intramuscularly twice with either 107 TCID50 rAdV-SFV-E2 or one dose of Bartha-K61 alone or 107 TCIDs0 rAdV-SFV-E2 combined with one dose of Bartha-K61 at an interval of three weeks, and challenged with the virulent CSFV Shimen strain at four weeks post-booster immunization. The results showed that all the pigs co-immunized with rAdV-SFV-E2 and Bartha-K61 developed high-level specific antibodies against CSFV and PRV comparable to either vaccine administered alone and fully protected the immunized pigs against lethal CSFV challenge. These data demonstrated that Bartha-K61 has no interference with the efficacy of rAdV-SFV-E2.
