摘要
为了解旋毛虫感染过程中血清和膈肌低氧诱导因子-1α(HIF-1α)的动态变化,本研究将旋毛虫ISS534株经口服感染90只大鼠,并于感染后不同时间各取6只大鼠收集血清和膈肌,采用荧光定量PCR和ELISA方法检测各时间点膈肌HIF-1α mRNA水平和血清HIF-1α水平。结果显示,旋毛虫发育成成虫至产生新生幼虫期间血清和膈肌HIF-1α含量无明显变化,产生新生幼虫后血清和膈肌HIF-1α水平逐渐升高,而且分别在6d和7 d时HIF-1α含量与感染前相比差异显著(p<0.05),随后呈逐渐上升趋势。血清HIF-1α含量直至9 d达到峰值并保持平稳状态,从14 d开始逐渐下降;而膈肌HIF-1α含量于9 d达到峰值后开始逐渐下降至肌幼虫形成,感染30 d血清和膈肌HIF-1α水平下降至感染前状态。结果表明,HIF-1α水平在旋毛虫感染过程中呈倒V型走势,HIF-1α可能是参与调控旋毛虫感染的重要因子。
To explore the dynamic changes of hypoxia-induced factor-1α (HIF-1α) in serum and diaphragm of rat infected with Trichinella spiralis (T.spiralis), serum and diaphragm in rats infected with T.spiralis ISS534 were collected at different days during a period of 30 days post-infection, respectively. The level of HIF-1α in serum and diaphragm were detected by ELISA method at different time points, and the mRNA expression of HIF-1α was detected using Real-time quantitative PCR. The level of HIF-1α in serum and diaphragm had no change during the stage of adult worms of T.spiralis. After newborn larvae were deposited by adult female worms, the level of HIF-1α in serum and diaphragm were increased gradually, and there was significant difference on the 6th day and 7th day of post-infection, respectively. And subsequently showed a tendency of gradual increase. The level of serum HIF-1α reached the highest level on the 9th day of post-infection and maintain stable, but decreased gradually began at the 14th day of post-infection. The level of diaphragm HIF-let was decreased gradually to ML forming after reached the highest level at the 9th day of post-infection. The level of HIF-let in serum and diaphragm were decreased to those pre infection at the 9th day of post-infection. The level of HIF-1α revealed the inverse V-shape distribution in rat infected with T.spiralis. HIF-1α may be an important regulatory factor in infection of T.spiralis.
