转染miR-200b基因的鼻咽癌CNE2细胞增殖、侵袭能力变化及其机制 被引量:1

摘要 目的观察转染miR-200b基因的鼻咽癌CNE2细胞增殖及侵袭能力变化,并探讨其机制。方法将鼻咽癌CNE2细胞分为观察组1、对照组1、观察组2、对照组2、观察组3。观察组1转染miR-200b模拟物,对照组1转染无关序列,观察组2转染BMI-1小干扰RNA(BMI-1 siRNA),对照组2转染RNA干扰无关序列,观察组3转染miR-200b抑制剂与BMI-1 siRNA。采用MTT实验观察细胞增殖能力,采用Transwell侵袭实验观察细胞侵袭能力。采用双荧光素酶报告基因检测系统检测BMI-1结合位点荧光素酶活性,Western blotting法检测BMI-1蛋白,qRTPCR检测BMI-1 mRNA。结果观察组1与对照组1相比,观察组2与对照组2相比,细胞48、72、96 h增殖能力减弱,(P均<0.05)。观察组1、对照组1、观察组2、对照组2、观察组3穿膜细胞数分别为(77.5±8.5)、(147.7±13.6)、(81.2±7.4)、(149.1±10.8)、(126.4±11.2)个;观察组1与对照组1相比,观察组2与对照组2相比,P均<0.01。miR-200b能与BMI-1非编码区结合,抑制BMI-1蛋白、mRNA的表达。结论转染miR-200b基因的鼻咽癌CNE2细胞增殖与侵袭能力降低,这可能是通过下调BMI-1实现的。
