HPV16-E7肽段鼻腔免疫联合淋巴毒素清除小鼠生殖道HPV16-E7慢病毒感染的作用

Effect of combined use of intranasal immune with HPV16-E7 lentivirus peptide and lymphotoxin injection on clearance of HPV16-E7 lentivirus infection in reproductive tract of rats

摘要 目的研究HPV16-E7肽段鼻腔免疫联合淋巴毒素(LT)清除小鼠生殖道HPV16-E7慢病毒感染的作用。方法建立HPV16-E7慢病毒感染模型。建模成功的小鼠24只随机均分为四组:A组实施HPV16-E7肽段鼻腔免疫,B组静脉注射LT,C组采用HPV16-E7鼻腔免疫+LT静脉注射,D组应用PBS作为对照。鼻腔免疫分别于建模后0、14和28d各进行1次。其后收集小鼠阴道黏膜组织及生殖道淋巴结,检测小鼠阴道组织中HPV16-E7蛋白的表达及淋巴结中淋巴归巢相关因子的表达情况。结果与B组和D组比较,C组及A组淋巴归巢因子在淋巴结中表达分别呈(■)及(+),HPV16-E7蛋白表达量下降(P<0.05)。结论 HPV16-E7肽段鼻腔免疫后,HPV16-E7慢病毒感染小鼠清除HPV16-E7的能力增加。LT能有效提高经鼻腔诱导的免疫效应,可能通过促进淋巴细胞的活化和调节淋巴归巢相关因子表达而增强免疫应答效应。 Objective To investigate the effect of combined use of intranasal immunization with HPV16-E7 lentivirus peptide and lymphotoxin injection on clearance of HPV16-E7 lentivirus infection in reproductive tract of rats. Methods Twenty-four mice models with HPV16-E7 lentivirus infection were established and then equally assigned into four groups of A(treated with intranasal immunization with HPV16-E7 lentivirus peptide), B(treated with intravenous injection of lymphotoxin),C(treated with intranasal immunization with HPV16-E7 lentivirus peptide plus lymphotoxin injection) and D (treated with PBS as the control). The intranasal immunization with HPV16-E7 lentivirus peptide was performed on the 0,14th and 28th day after model establishment. The vaginal mucosa and genital lymph nodes were collected for detecting the expressions of HPV16-E7 protein and the factors related to lymphatic homing. Results Compared to groups of B and D, the expressions of the factors related to lymphatic homing nodes were as (++) and (+), and HPV16-E7 protein expression was lower (P^0. 05). Conclusion The ability of the HPV-infected cell clearance is increased in the mice infected with HPV16-E7 virus after intranasal HPV16-E7 immunization, which is effectively improved by LT possibly through promoting lymphocyte activation and regulation of lymphatic homing related factors.
