紫苏甾醇超声波辅助提取工艺及其抗氧化性研究 被引量:9

摘要 研究紫苏甾醇超声波辅助提取工艺及其抗氧化性。在单因素试验基础上,选择超声温度、超声时间和液料比分析其对紫苏甾醇提取效果的影响,设计L_9(3~4)正交试验对超声波辅助提取工艺进行优化。采用半数抑制率(IC_(50))表示法对9个不同紫苏品种甾醇提取物的抗氧化性进行比较。结果表明,紫苏甾醇超声波(200 W,40 k Hz)辅助提取最佳工艺条件为:液料比4∶1,超声时间50min,超声温度50℃。在最佳工艺条件下,紫苏甾醇得率可达2.604 mg/g。不同品种紫苏甾醇提取物抗氧化性差异极显著,其中ZB-1在9个品种中甾醇含量最高,对DPPH·和·OH的清除能力最强,O^-_2·的清除能力仅次于ZY-7。研究结果可从提取工艺和优质品种资源筛选两个方面为紫苏开发利用提供理论依据和技术参考。 Ultrasound - assisted extraction process and antioxidant activity of phytosterol from perilla were studied. Based on single factor experiment, the effects of ultrasonic temperature, ultrasonic time and ratio of liquid to solid on the extraction result of phytosterol from periUa were researched, and the ultrasound - assisted extraction process was optimized by L9 (3^4 ) orthogonal experiment. The antioxidant activities of phytosterols extracted from nine different varieties of perilla were compared by half inhibition rate (IC50 value) method. The results showed that the optimal ultrasound - assisted (200 W, 40 kHz) extraction conditions of phytosterol from perilla were obtained as follows :ratio of liquid to solid 4: 1, ultrasonic time 50 min, ultrasonic temperature 50 ℃. Under these conditions, the yield of phytosterol from perilla was 2. 604 mg/g. There were significant differences in the antioxidant activities of phytosterol extracted from different varieties of perilla. The content of phytosterol in ZB - 1 was the highest among the nine varieties. The scavenging capacities of ZB - 1 on DPPH· and · OH were the strongest, and its scavenging capacityon O2- ·was only weaker than that of ZY - 7. The study results could provide theoretical and technical references from the aspects of extraction process and screening high quality varieties re- sources for the development and utilization of pc- rilla.
