摘要
用传统培养法从广西梧州六堡茶中初筛获得5株真菌菌株,再用烟草梗丝培养基复筛,得到一株可以高效降解梗丝果胶和纤维素的产香菌菌株,经ITS及部分r DNA序列分析及形态特征观察,将其鉴定为塔宾曲霉(Aspergillus tubingensis),命名为GYC 501。该菌株在发酵培养过程中,能够产生令人愉悦的特殊香气,并且具有很好的果胶酶和纤维素酶活性,在2%蔗糖察氏液体培养基中培养36 h,纤维素酶活力达到418.2 U,培养48 h,果胶酶活力达到471.4 U。该菌株有望用于烟草梗丝发酵,降解梗丝中的果胶和纤维素,提高梗丝的质量和工业可用性。
Five fungi were isolated from Guangxi Wuzhou Liu Bao Tea by the method of traditional culture, the second screening was done by the culturing in the tobacco stem medium, an aroma-producing fungus which was effective in the degration of pectin and cellulose was acquired, and identified as A spergillus tub inge nsis based on ITS and partial rDNA sequence and morphology analysis, named as GYC 501. In the process of fermentation, this strain produces amiable aroma and had high yield of enzyme activity both for pectinase and cellulase. Culturing in Czapek-Dox medium with 2% sucrose, inoculated for 36 h, the cellulase activity was the highest, reached 418.2 U; inoculated for 48 h, the pectinase activity was the highest at 471.4 U. This strain was supposed to be used in degrading pectin and cellulose and fermentation of tobacco stem to reduce pectin and cellulose, then improved the quality of tobacco stem and its industrial availability.
