不同保存方法对柽柳基因组总DNA产量和质量的影响 被引量:5

Effects of Different Preservation Methods on Yield and Quality of Total Genomic DNA of Tamarix chinensis

摘要 通过比较不同保存方法对柽柳(Tamarix chinensis)基因组总DNA提取效果的影响,确定合适的野外采集植物样品的保存方法。采用改良CTAB法提取了室温风干、4℃、-20℃、-80℃和硅胶干燥5种保存方法分别保存24 h、1周和1月后的柽柳基因组总DNA,结果表明:4℃保存1月后DNA产量最低,为60.35μg/g,-80℃条件下保存24 h的材料中提取的DNA产量最高,为246.92μg/g,保存效果最好。但在野外仪器条件不具备的情况下,柽柳可采用室温风干或硅胶干燥保存的方法,1月内材料中DNA降解程度较轻,可以扩增出所需目的片段。 Through the comparison of different preservation methods in Tamarix chinensis genome DNA, and determine the appropriate preservation method of wild-collected plant samples. The modified CTAB method was used to extract the genome DNA of Tamarix chinensis at room temperature, 4℃, -20℃, -80℃, and silica drying with each treatment for 24 hours, one week, one month, respectively. The results showed that DNA yield was the lowest (60.35 txg/g) after 1 month preservation under the condition of 4℃, and DNA yield was the highest (246.92 μg/g) after 24 h preservation under the condition of-80℃. However, in the wild, when the instruments were not available, Tamarix chinensis can be preserved at room temperature or silica-gel, DNA degradation of materials was slight within 1 month, which can be used for amplify the desired fragment.
