摘要
肿瘤细胞和正常组织细胞具有许多相似的细胞表型特征,因此如何增强治疗的靶向性极为重要。沃伯格效应是指肿瘤细胞在有氧条件下仍依赖糖酵解,而2-脱氧-D-葡萄糖(2DG)是最常用的糖酵解抑制剂,能特异性地抑制恶性细胞增殖、侵袭、迁移和黏附并增强免疫原性。由于单独应用2DG所需剂量较大使其在临床推广受限,故现研究多着手于2DG与传统化疗药物的联用或与其他代谢抑制剂及新药合用。
Malignant cells share many similar phenotype characteristics with normal tissue cells,there- fore, it is extremely critical to improve the target activity of anticancer agents. Warburg effect refers that tumor cells still rely on glycolysis under aerobic condition, and 2-deoxy-D-glucose (2DG) is the most common gly- colysis inhibitor, which can specificly restrain the proliferation, invasion, metastasis, adhesion of malignant cells and enhance the immunogenicity of malignant ceils. Due to the megadose needed when applying 2DG alone, the clinical promotion is restricted. Therefore, currently the studies are turning to the combination of traditional chemotherapy drugs, other metabolic inhibitors or new drugs with 2DG.
