不同固定液对内皮细胞形态和EP4荧光染色的影响 被引量:3

Effect of different fixative on endothelial cells morphology and immunostaining of EP4 receptor

摘要 采用4种固定液对人脐静脉内皮细胞(HUVECs)进行固定:1-20℃无水甲醇溶液;2新鲜配制4 g/L多聚甲醛溶液;3久置(≥1个月)4 g/L多聚甲醛溶液;4 3.7%甲醛溶液。观察细胞形态和前列腺素受体4(EP4)免疫荧光染色效果。结果表明3.7%甲醛溶液固定后进行免疫荧光染色显示大部分细胞维持梭形或不规则多边形,EP4荧光染色效果较佳;所以3.7%甲醛溶液固定液固定细胞20 min,HUVECs形态典型,EP4的荧光染色效果最佳。 Human umbilical endothelial cells( HUVECs) were fixed by four different fixative,1 cells were fixed with precooled methanolsolution(-20℃); 2 cells were fixed with fresh preparation of 4 g / L paraformaldehyde solution( PFA); 3 cells were fixed with old 4 g / L PFA solution( more than a month); 4 cells were fixed with3. 7% formaldehydesolution. Cells which were fixed with 3. 7% formaldehydesolution maintained fusiform or irregular polygon,and the prostaglandin recepter 4( EP4) immunostaining effect was the best. We concluded that 3. 7%formaldehydesolution for 20 min was suitable for HUVECs immunofluorescence,HUVECs maintain typical cell morphology and the EP4 immunostaining effect was the best.
