皮肤肿瘤患者不同组织中PinX1人端粒酶逆转录酶mRNA的表达意义 被引量:3

摘要 目的探讨基底细胞癌(BCC)、鳞状细胞癌(SCC)和日光性角化病(AK)组织中PinX1蛋白和人端粒酶逆转录酶(hTERT)mRNA的表达及其意义。方法选取2010年3月至2014年9月间在西安交通大学第一附属医院就诊的49例皮肤肿瘤患者作为观察组,其中BCC患者16例、SCC患者18例、AK患者15例。另选取15例正常人皮肤组织作为对照组。采用半定量逆转录聚合酶链反应(RT-PCR)法,检测BCC、SCC及AK患者组织和15例正常人皮肤组织中PinX1和hTERT的mRNA表达水平,并检测其中的端粒酶活性。结果 PinX1 mRNA在3种不同皮肤肿瘤组织中的相对表达量较正常对照组低,差异有统计学意义(P<0.05),但在3种不同皮肤肿瘤组织之间的相对表达量比较差异无统计学意义(P>0.05)。hTERT mRNA在3种不同皮肤肿瘤组织中的相对表达量较正常对照组高,差异有统计学意义(P<0.05),但在3种不同皮肤肿瘤组织之间的相对表达量比较差异无统计学意义(P>0.05)。结论 PinX1的下调及hTERT上调可能与皮肤肿瘤形成过程中端粒酶激活性及维持机制有关,二者在基底细胞癌、鳞状细胞癌细胞凋亡方面可能存在拮抗作用,检测PinX1和hTERT的表达水平,对评价皮肤肿瘤预后有重要的临床价值。 Objective To investigate PinX1,h TERT mRNA expression and its significance in basal cell carcinoma,squamous cell carcinoma,actinic keratosis tissues. Methods Forty-nine patients were selected between March 2010 to September 2014 of skin cancer patients between March 2010 to September2014 in our hospital,where 16 patients with BCC,SCC,18 patients and 15 cases of AK patients,as the observation group; selected 15 cases of normal human skin tissue as in the control group. At the same time semi-quantitative reverse transcription polymerase chain reaction( reverse transcription-polymerase chain reaction,RT-PCR) to detect XX cases of basal cell carcinoma,squamous cell carcinoma,actinic keratosis tissue and normal skin tissue PinX1,h TERT mRNA expression levels and telomerase activity in detecting them. Results BCC,SCC and AK tissue PinX1 mRNA relative expression levels lower than the normal control group,there was significantdifference between two groups.( P〈0. 05),the relative expression of difference BCC and SCC and AK tissue PinX1 mRNA was not statistically significant( P〈0. 05). BCC,the relative expression levels of SCC and AK tissues h TERT mRNA was higher than the normal control group,there was significantdifference between two groups.( P〈0. 05),the relative expression of difference BCC and SCC and AK tissues h TERT mRNA was not statistically significant( P〉0. 05). Conclusions PinX1 h TERT upregulation and downregulation may be associated with skin tumor formation and maintenance of telomerase activation mechanisms,both in the basal cell carcinoma,squamous cell antagonism may exist Apoptosis aspect. Detection of expression level of h TERT PinX1 and evaluation of skin cancer prognosis has important clinical value.
