HJURP基因表达下调对人胚胎绒毛细胞增殖凋亡作用的研究

摘要 目的研究HJURP基因的表达下调对人胚胎绒毛细胞增殖、细胞周期和细胞凋亡的影响。方法用CCK8法、Annexin V-EGFP/PI双染法、TUNEL法、Trans Well迁移侵袭实验等方法研究HJURP表达下调对人胚胎绒毛细胞增殖、细胞凋亡及细胞周期的影响。结果 HUJRP基因表达下调后,与对照组比较,明显抑制胚胎绒毛细胞的增殖及迁移侵袭能力。胚胎绒毛细胞的凋亡率为9.87±1.92%增加至,明显高于NS组3.92±1.12%(P<0.01)。对细胞周期的影响表现为G0/G1期阻滞,G0/G1期细胞从42.9±3.12%增加至62.4±1.92%。Trans Well侵袭实验结果显示,实验组穿过Trans Well膜的数目明显少于对照组。结论 HJURP基因的表达下调可以导致人胚胎绒毛细胞增殖抑制,诱导人胚胎绒毛细胞凋亡,使人胚胎绒毛细胞周期阻滞于G0/G1期,推测HJURP基因表达下调与人胚胎自然流产有相关性。 Objective:To study effect of silencing HJURP gene on proliferation,cycle and apoptosis of human embryonic villus cells,to study the possible mechanism of HJURP gene in spontaneous abortions. Methods:The effect of silencing HJURP gene were investigated by the methods of CCK8,Annexin V-EGFP / PI double staining,TUNEL method,Trans Well migration assay in human embryonic villus. Results:Compared with the control group,silencing HJURP gene significantly inhibited the activity of embryonic villus cells and cell cycle G2 / M phase,inhibited the proliferation and migration and invasion capacity of the cells. And TUNEL-positive rate was significantly higher,and improved the level of apoptosis. Conclusion:Silencing HJURP gene can induce apoptosis and cell cycle arreste in G0 / G1 phase,its action mechanism may be correlated with the occurrence of spontaneous abortion.
