金水宝胶囊联合二甲双胍对糖尿病勃起功能障碍大鼠糖代谢、AGEs及AngⅡ的实验研究

Effect of Jinshuibao Capsuleand melbine on blood glucose,AGEs and AngⅡ levels in rats with diabetic erectile dysfunction

目的:观察金水宝胶囊对糖尿病勃起功能障碍大鼠血糖、糖化血红蛋白、晚期糖基化终产物(AGEs)及血管紧张素Ⅱ(AngⅡ)的影响。方法:采用SD大鼠腹腔注射链脲佐菌素(STZ),造模后第3d、第1周、第2周断尾取血检测血糖,当血糖大于16.6 mmol/L时同时颈项皮肤松软处注射阿朴吗啡无勃起,提示糖尿病勃起功能障碍大鼠模型造模成功。将动物随即分为金水宝高剂量治疗组、金水宝中剂量治疗组、金水宝低剂量治疗组、阳性对照组、模型组各10只,各组间大鼠体重、血糖无显著性差异。同时留10只作为空白对照组。给药8周后测定大鼠血糖浓度、糖化血红蛋白含量、AGEs及AngⅡ水平。结果:与模型组相比,金水宝高、中、低剂量治疗组均能够降低血糖、糖化血红蛋白、AGEs及AngⅡ水平(P<0.01),但以金水宝高剂量组最显著(P<0.01),其中血糖监测中,金水宝中、低剂量组治疗组之间无意义,糖化血红蛋白中,金水宝低剂量治疗组与阳性对照组之间无意义。AGEs及AngⅡ水平的金水宝中、低剂量治疗组之间无统计学意义(P>0.05)。结论:金水宝能有效协助降低血糖、糖化血红蛋白、AGEs及AngⅡ水平,其中金水宝高剂量治疗组疗效最显著,为该制剂临床用于治疗糖尿病勃起功能障碍提供了实验理论依据。

Objectives： To observe the influence of Jinshuibao Capsule and melbine on blood glucose,GHb,AGEs and Ang II levels in rats with diabetic erectile dysfunction. Methods： Rats were injected with intraperitoneal STZ to establish their DED model and randomly divided into five groups： control group,model group and Jinshuibao capsule high / medium / low dose group. The fasting blood glucose concentration,glycated hemoglobin levels and advanced glycation end- products and angiotensin of rats were determined in 8 weeks later. Results：Compared with model group,Jinshuibao capsule high,medium and low dose group and melbine can decrease the AngII level（ P〈0. 01）,the most significant in high dose group（ P〈0. 01）. Medium and low dose group had no statistically significant difference in blood glucose,FPG and AGEs and AngII（ P〉0. 05）; low dose group and control group had no statistically significant difference in glycosylated hemoglobin（ P〉0. 05）. Conclusions： Jinshuibao Capsule can reduce blood glucose and glycated hemoglobin level,AGEs and AngII expression,especially used in high dose,which provides basis for clinical treatment of diabetic neuropathy.
