摘要
目的探讨表皮生长因子(EGF)对角质形成细胞(HaCaT细胞)miR-21/PDCD4表达的调控作用。方法聚合酶链反应(PCR)法检测不同浓度EGF(0、5、15、25 ng/mL)对HaCaT细胞miR-21表达的影响;蛋白免疫印迹法检测不同浓度EGF(0、5、15、25 ng/mL)对HaCaT细胞PDCD4蛋白表达的影响。结果 EGF浓度依赖性促进HaCaT细胞miR-21的表达,与对照组比较,5 ng/mL EGF组、15 ng/mL EGF组、25 ng/mL EGF组HaCaT细胞miR-21表达分别为1.46±0.11、2.44±0.05、3.19±0.08(P<0.05);EGF浓度依赖性抑制HaCaT细胞HaCaT细胞PDCD4表达,跟对照组比较,25 ng/mL EGF组能显著抑制HaCaT细胞PDCD4表达。结论 EGF促进HaCaT细胞miR-21的表达,抑制HaCaT细胞PDCD4的表达水平。
Objective In order to explore the effects of epidermal growth factor (EGF) on the expression of miR-21/PDCD4 in HaCaT cells. Methods Polymerase Chain reaction (PCR) was used to detect the effect of different concentrations of EGF (0, 5, 15, 25 ng/mL) group on the expression of miR-21 in HaCaT cells. Western blotting was used to detect the expression of PDCD4 in HaCaT cells reduced by different concentrations of EGF (0, 5, 15, 25 ng/mL). Results EGF promoted the expression of miR-21 with concentration dependent manner in HaCaT cell. Comparing with the control group, the expressions of miR-21 in 5, 15, 25 ng/mL EGF treated groups were elevated 1.46±0.11, 2.44±0.05, 3.19±0.08 times (P〈0.05). EGF inhibited the expression of PDCD4 with concentration dependence manner in HaCaT cell. Comparing with the control group, the expression of PDCD4 in HaCaT cell treated with 25 ng/mL EGF was significantly inhibited. Conclusion EGF promotes the expression of miR-21 and inhibits the PDCD4 expression level in HaCaT cell.
