摘要
目的观察槐耳颗粒对单侧输尿管梗阻模型(UUO)肾组织beclin-1、FN的影响来进一步阐明槐耳在急性肾损伤中作用的可能机制。方法 Wistar大鼠随机分为假手术组(Shan组),模型组(UUO组),模型组+槐耳治疗组(UHE组),造模后0、3、7天分析大鼠肾组织病理损伤,血生化Scr、Urea值,beclin-1、FN的表达分布及变化。结果 1造模后7天,UUO组较Sham组病理损伤评分和血生化指标Scr、Urea均有显著上调(P<0.05)。2造模后3天、7天,UUO组beclin-1、FN在肾小管上皮细胞中表达上调(P<0.05)。3造模后3天、7天,槐耳治疗组较模型组Scr、Urea、beclin-1、FN均有显著下调(P<0.05)。结论梗阻性肾损伤中,槐耳可以抑制肾组织中自噬的发生,从而减缓小管间质纤维化的形成,延缓小管间质的损伤进程。
Abstract:Objective This study was designed to explore the mechanism of Huaier(HE) effects on acute kidney injury, by in- vestigating the variation of beclin- 1 and FN in unilateral ureteral obstruction (UUO) with rat. Methods Wistar rats were ran- domly divided into Sham, UUO, UHE(UUO + HE) groups. After administration of HE for 0, 3, 7 days, the levels of Set, Urea and the distribution of the beclin - 1 and FN were measured by biomedical filters and western blot/immunohistoehemistry. Results After HE administration, Scr, Urea, beclin - 1 and FN were decreased in all time points (P 〈 0.05 ). The level of beclin - 1 and FN were increased in all time points in UUO rats, but the pathological scores, Scr and Urea were only increased at day 7. Con- clusion HE could defer tubulointerstitial injury in obstructive nephropathy by inhibiting autophagy to ameliorate tubulointerstitial fibrosis.
